Article Abstract

Tocopheryl succinates (TOSs) are, in contrast to tocopherols, highly cytotoxic against many cancer cells. In this study the enzyme activity of secretory phospholipase A(2) towards various succinate-phospholipid conjugates has been investigated. The synthesis of six novel phospholipids is described, including two TOS phospholipids conjugates. The studies revealed that the TOS conjugates are poor substrates for the enzyme whereas the phospholipids with alkyl and phenyl succinate moieties were hydrolyzed by the enzyme to a high extent.

As part of our investigations into C-C bond scission and functionalization, we report a halodealkenylation in which the C(sp)-C(sp) bonds of alkenes are cleaved and C(sp)-halide bonds are formed, via a radical intermediate. These transformations occur through Criegee ozonolysis and Fe-catalyzed reductive coupling assisted by vitamin C as a stoichiometric reductant. We applied this strategy to the formal synthesis of ()-γ-tocopherol.

This study compares the effects of vitamins - including ascorbic acid, its derivative 3-O-ethyl-ascorbic acid (EAA), and tocopherol - as well as the main non-psychoactive phytocannabinoids on the viability of various skin cells, including healthy (keratinocytes/melanocytes/fibroblasts) and cancer cells (melanoma/SCC), under standard culture conditions and after the exposure to UVA radiation. All the conducted tests (MTT, SRB, and LDH) consistently indicate that the regenerative effect of EAA is stronger than that of ascorbic acid, while tocopherol acts selectively on healthy/cancer cells, inducing or inhibiting their proliferation, respectively. In the case of phytocannabinoids, only cannabidiol shows protective/regenerative properties for healthy cells.
