Aims: The aim of the present study is to evaluate the effects of baicalein on the 6-hydroxydopamine (6-OHDA)-induced rat model of Parkinsonian tremor and elucidate the potential mechanism.

Main Methods: Adult male Sprague-Dawley rats were subjected to Parkinsonian tremor by 6-OHDA-medial forebrain bundle (MFB) injection. Baicalein (100, 200 and 400mg/kg) was administrated by gavage once daily, for 1 week. The anti-tremor effect of baicalein on a model of Parkinsonian tremor was examined. Dopamine (DA), glutamate (GLu) and gamma-aminobutyric acid (GABA) levels in basal ganglia were determined by HPLC-ECD. Cytochrome oxidase subunit I (COI) mRNA expression in subthalamic nucleus (STN) was assessed by real-time RT-PCR. GABA transaminase (GABA-T) and glutamine synthetase (GS) protein expression in basal ganglia were tested by immunostaining. Effects of baicalein on [Ca(2+)]i induced by GLu were tested in primary cultured mesencephalic neurons.

Key Findings: Baicalein significantly attenuated muscle tremor of PD rats. The metabolic balance between GLu and GABA was modulated by baicalein treatment. It was found that treatment with baicalein inhibited the STN's COI mRNA expression in experimental PD. In rat primary mesencephalic cultures, baicalein inhibited significantly GLu-induced intracellular calcium [Ca(2+)]i increase.

Significance: These results suggest that baicalein plays a neuromodulatory role in balancing GABA and GLu neurotransmitter in basal ganglia, and might be a promising candidate for the treatment of tremor-dominant type of idiopathic Parkinson's disease.

L-type calcium channel antagonists are uncommon causes of myoclonus, and the underlying mechanism remains unclear. Here, we report a case of parkinsonian syndrome with deterioration of preexisting myoclonus after nifedipine use. A 96-year-old woman was administered a single dose of sustained-release nifedipine for chest pain.

Parkinson's disease (PD) is a prevalent neurodegenerative disorder characterized by the degeneration of dopamine neurons in the substantia nigra pars compacta, leading to motor and non-motor symptoms. While motor symptoms such as rigidity, tremor, bradykinesia/akinesia, and postural instability are well-recognized, non-motor symptoms including cognitive decline, depression, and anxiety also significantly impact patients' quality of life. Preclinical research utilizing animal models has been instrumental in understanding PD pathophysiology and exploring therapeutic interventions.

Differences in Blood and Cerebrospinal Fluid Between Parkinson's Disease and Related Diseases.

It is difficult to distinguish Parkinson's disease (PD) in the early stage from those of various disorders including atypical Parkinson's syndrome (APS), vascular parkinsonism (VP), and even essential tremor (ET), because of the overlap of symptoms. Other, more challenging problems will arise when Parkinson's disease develops into Parkinson's disease dementia (PDD) in the middle and late stages. At this time, the differential diagnosis of PDD and DLB becomes thorny.

Spasmodic dysphonia: the need for a combined neurological and phoniatric approach.

Spasmodic dysphonia (SD) is now generally considered to be a task-specific focal dystonia. For the first time, we wanted to explore the relationship between SD and dystonia from a combined neurological and phoniatric perspective. For this, we studied 115 patients with non-psychogenic SD by a combined neurological and phoniatric evaluation.

Surface Electromyography for the Diagnosis of Tremor Syndrome: A Study of 97 Patients.

Department of Neurology and Stroke Medicine, Amrita Hospital, Faridabad, Delhi National Capital Region, India.

Article Synopsis
  • Tremor is a common movement disorder with various causes, and surface electromyography (SEMG) is an effective tool for diagnosing different tremor syndromes.
  • A study of 97 patients at a movement disorder clinic revealed that essential tremor was the most prevalent syndrome, with distinct electrophysiologic patterns observed across different types of tremors.
  • The results indicate that SEMG can help distinguish between tremor-ataxia syndrome and essential tremor, which can be challenging to differentiate in clinical practice.
