Advances in analysis of human milk oligosaccharides.

Oligosaccharides in human milk strongly influence the composition of the gut microflora of neonates. Because it is now clear that the microflora play important roles in the development of the infant immune system, human milk oligosaccharides (HMO) are studied frequently. Milk samples contain complex mixtures of HMO, usually comprising several isomeric structures that can be either linear or branched. Traditionally, HMO profiling was performed using HPLC with fluorescence or UV detection. By using porous graphitic carbon liquid chromatography MS, it is now possible to separate and identify most of the isomers, facilitating linkage-specific analysis. Matrix-assisted laser desorption ionization time-of-flight analysis allows fast profiling, but does not allow isomer separation. Novel MS fragmentation techniques have facilitated structural characterization of HMO that are present at lower concentrations. These techniques now facilitate more accurate studies of HMO consumption as well as Lewis blood group determinations.

Preparation of mixed-mode hydrophilic particle for efficient separation of common human milk oligosaccharides.

Human milk oligosaccharides (HMOs) are essential for babies' growth. The most commonly used method for HMOs analysis combines hydrophilic interaction liquid chromatography (HILIC) with ultra-performance liquid chromatography (UPLC). However, high-pressure operation and the cost of commercial HILIC columns limit progress in the HMOs research.

Lactoferrin (Lf) is a multifunctional iron-binding glycoprotein, involved in a wide range of bioactivities, including immunomodulatory and antiviral activities. Lf in human milk and bovine Lf added to infant formula may provide some protection against viral infections. However, functions of Lfs from different sources may differ due to varying manufacturing processes and posttranslational modifications.

Microglia mediate the early-life programming of adult glucose control.

Glucose homeostasis is, in part, nutritionally programmed during early neonatal life, a critical window for synapse formation between hypothalamic glucoregulatory centers. Although microglia prune synapses throughout the brain, their role in refining hypothalamic glucoregulatory circuits remains unclear. Here, we show that the phagocytic activity of microglia in the mediobasal hypothalamus (MBH) is induced following birth, regresses upon weaning from maternal milk, and is exacerbated by feeding dams a high-fat diet while lactating.

Probiotic Bifidobacterium bifidum strains desialylate MUC13 and increase intestinal epithelial barrier function.

Sci Rep

Department of Biomolecular Health Sciences, Division Infectious Diseases and Immunology, Faculty of Veterinary Medicine, Utrecht University, Utrecht, The Netherlands.

Probiotic bacteria including Bifidobacterial species have the capacity to improve intestinal health, but the underlying molecular mechanisms are often not understood. Bifidobacteria are considered keystone species but have a relatively low abundance in the adult intestinal tract. Bifidobacterium colonization depends on degradation of host-derived carbohydrates, including human milk oligosaccharides and mucin-associated oligosaccharides.

[«PFAS», pollutants in the spotlight].

Rev Med Liege

Service de Toxicologie Clinique, Médico-légale, de l'Environnement et en Entreprise. CHU Liège, Belgique.

In November 2023, Wallonia was rocked by the discovery of problematic PFAS contamination of drinking water in the Chièvres region. PFAS («per- or polyfluoroalkyl substances») are compounds produced and used in large quantities by human activities for numerous applications, such as the formulation of fire-fighting foams, which appears to be at the origin of the pollution in Chièvres. The two main PFASs (perfluorooctanoic acid (PFOA) and perfluorooctane sulfonic acid (PFOS)) have been shown to have toxic effects on immunity, cholesterol metabolism, liver function and newborn birth weight.
