Synthesis of N-substituted acridinediones and polyhydroquinoline derivatives in refluxing water.

Acridinediones were synthesized by the one-pot Hantzsch condensation of an aromatic aldehyde, 5,5-dimethyl-1,3-cyclohexanedione, and aniline/4-methylaniline in refluxing water. This method has then been extended to the four-component reaction of an aromatic aldehyde, 5,5-dimethyl-1,3-cyclohexanedione, ethyl acetoacetate and ammonium acetate for the synthesis of polyhydroquinoline derivatives. This is an environmentally friendly and efficient procedure providing  good to excellent yields.

In the present report, we have described the synthesis of N-aminopolyhydroquinoline (N-PHQ) derivatives using highly efficient -cyclodextrin (-CD) as a catalyst by the Hantzsch condensation of substituted aromatic aldehydes, dimedone, and hydrazine hydrate in one pot. The reactions were completed in a shorter time without the generation of any other byproduct. The synthesized N-PHQs were washed thoroughly with distilled water and recrystallized with ethanol to get highly purified products (as crystals).

These molecules were characterized by modern spectroscopic techniques (H-NMR, C NMR, and LC-HRMS) and their antibacterial and α-glucosidase inhibitory activities were assessed. Compound 8 was found to be the most active inhibitor against NCTC 5348, IM 622, , and ATCC 6337 with a zone of inhibition of 15.
