Copper-catalyzed cycloaddition of steroidal azides and ferrocenyl-alkynes were found to be an efficient methodology for the synthesis of ferrocene-labeled steroids. At the same time, a great difference between the reactivity of 2β- or 16β-azido-androstanes and a sterically hindered 6β-azido steroid toward both ferrocenyl-alkynes and simple alkynes, such as phenylacetylene, 1-octyne, propargyl acetate and methyl propiolate, was observed.

Nanoelectrodes, renowned for their small size, rapid mass transport, fast response, and high spatiotemporal resolution, have been recognized as a powerful tool in biosensing, especially for single-cell analysis. However, the nanoelectrode itself has no selectivity and cannot respond to nonelectroactive substances, limiting its wide application to some extent. Herein, we propose a simple and efficient electrochemical conjugation strategy to develop an electrochemical aptamer-coupled (E-AC) sensor for detecting adenosine triphosphate (ATP) in single living cells.

Ferrocene (Fc)-based disulfide molecules of various lengths with amino acid scaffolds and alkane or oligo(phenylene-ethynylene) (OPE) bridges are used in a mixed SAM with a di-(ethylene oxide) terminal mercaptoundecanol diluent (PEG). The relative height of the Fc redox reporter in the SAM is compared to determine if there are protective effects like antifouling and specific detection. The HaloTag-binding motif is used as a proof-of-concept to investigate the electrochemical response to the HaloTag protein due to its known covalent and fast linkage.
