Introduction: Non-invasive prenatal diagnosis (NIPD) of Down syndrome is rapidly evolving. Currently, two applications for NIPD of Down syndrome have been developed with potential and have displayed positive results; the NIPD using next-generation sequencing technologies and the NIPD using the methylated DNA immunoprecipitation (MeDIP) real-time quantitative polymerase chain reaction (qPCR).

Areas Covered: The MeDIP real-time qPCR approach is based on the identification of differentially methylated regions (DMRs) and their use for discriminating normal from Down syndrome cases. DMRs were identified using high-resolution oligo-arrays. A subgroup of DMRs was selected for further investigation. Through the design of a discriminant equation which combines the results obtained from different DMRs, normal and abnormal cases are correctly classified indicating 100% sensitivity and specificity.

Expert Opinion: Previous studies have also identified DMRs between non-pregnant female blood and placental DNA. However, these methods have been associated with a number of limitations including the low sensitivity and/or specificity of the assays, the limited number of identified DMRs or methylation sensitive sites and SNPs located on DMRs. These limitations have been overawed by the development of the MeDIP real-time qPCR-based methodology.

Objective: To investigate the effect of epigenetic regulation on spermatogenic dysfunction-related infertility (SDI) in C57BL/6J male mice.

Methods: Sixty C57BL/6J male mice were randomly divided into a normal control and an SDI model group and the SDI model was established using the epididymis-targeting polypeptide CSA combined with indocyanine green-loaded free nanoparticles (ICG-NPS), busufan and dimethyl sulfoxide (DSMO). After intervention with 5-AZA-DC, the epididymides were collected from the mice for measurement of the rates of sperm DNA fragmentation (SDF), sperm acrosome integrity (SAI) and spontaneous acrosome reaction (SAR), amplification of the ERp29 gene by FISH, determination of the mRNA and protein expressions of DNMT1, ERp29, PTEN and TSC2 by quantitative real-time PCR and Western blot, and analysis of the ERp29, PTEN and TSC2 genes by methylated DNA immunoprecipitation sequencing (MeDIP-seq).

Young Breast Cancer: Novel Gene Methylation in WBC.

Introduction: Breast cancer is a highly diverse disease, and epigenomic alterations, as principle changes in the pathogenesis of breast cancer, have recently been noticed in epimarker research on peripheral blood.

Methods: In this study, DNA samples isolated from the white blood cells of 30 breast cancer patients were compared to 30 healthy controls using methylated DNA immunoprecipitation microarray (MeDIP-chip) to determine differentially methylated region as a potential epimarker in cancer and control cases.

Results: A total of 1799 differentially methylated regions were identified, including ZNF154, BCL9, and HOXD9, in which significant methylation differences were confirmed in breast cancer patients through a quantitative real-time polymerase chain reaction.
