The purpose of this case report is to demonstrate the effectiveness of a matrix consisting of recombinant human platelet-derived growth factor BB (rhPDGF-BB)-hydrated cancellous allogenic block graft in the reconstruction of large local human alveolar ridge defects. The results suggest improved bone regeneration when combining rhPDGF-BB with the allogenic block graft. The clinical and histologic evidence of new bone formation as well as bone remodeling supports the clinical potency of this growth factor-mediated therapy.

Signaling through the platelet-derived growth factor receptor alpha (PDGFRα) plays a critical role in craniofacial development. Phosphatidylinositol 3-kinase (PI3K)/Akt is the primary effector of PDGFRα signaling during mouse skeletal development. We previously demonstrated that Akt phosphorylates the RNA-binding protein serine/arginine-rich splicing factor 3 (Srsf3) downstream of PI3K-mediated PDGFRα signaling in mouse embryonic palatal mesenchyme (MEPM) cells, leading to its nuclear translocation.

Aim: The study aimed to evaluate the effects of alpha hydroxy acids and chelating agents on dentin conditioning for the release of growth factors.
Methods: The agents used for dentin conditioning included 17% ethylenediaminetetraacetic acid (EDTA), 10% glycolic acid (GA), 10% citric acid (CA), and 5% maleic acid (MA). Forty horizontally sectioned (SV1) human dentine slices were conditioned for 5 and 10 minutes so that the growth factor liberation reached quantifiable levels.
