Published: November 1990

Redundancy of the transposed thoracic colon is not uncommon. Rarely, however, when accompanied by severe symptoms, it may necessitate surgical correction. We describe herein a surgical technique that allows the stretching and straightening of the redundant transposed organ.

The number of species with high-quality genome sequences continues to increase, in part due to the scaling up of multiple large-scale biodiversity sequencing projects. While the need to annotate genic sequences in these genomes is widely acknowledged, the parallel need to annotate transposable element (TE) sequences that have been shown to alter genome architecture, rewire gene regulatory networks, and contribute to the evolution of host traits is becoming ever more evident. However, accurate genome-wide annotation of TE sequences is still technically challenging.

Article Synopsis
  • PIWI-interacting RNAs (piRNAs) help maintain genomic integrity and reproductive function by regulating transposable elements and protein-coding genes in germline cells.
  • Recent findings in golden hamsters indicate that piRNAs play a critical role in female fertility, highlighting limitations in the mouse model for studying the PIWI-piRNA pathway.
  • The review discusses advancements in piRNA research, its impact on female fertility, and future directions for understanding reproductive mechanisms in mammals.

The Enterobacteriaceae are a scientifically and medically important clade of bacteria, containing the model organism , as well as major human pathogens including and . Essential gene sets have been determined for several members of the Enterobacteriaceae, with the Keio single-gene deletion library often regarded as a gold standard. However, it remains unclear how gene essentiality varies between related strains and species.

Genetic interactions identify functional connections between genes and pathways, establishing gene functions or druggable targets. Here we use CRISPRi-TnSeq, CRISPRi-mediated knockdown of essential genes alongside TnSeq-mediated knockout of non-essential genes, to map genome-wide interactions between essential and non-essential genes in Streptococcus pneumoniae. Transposon-mutant libraries constructed in 13 CRISPRi strains enabled screening of ~24,000 gene pairs.
