The European Union's ban on animal testing for cosmetic products and their ingredients, combined with the lack of validated animal-free methods, poses challenges in evaluating their potential repeated-dose organ toxicity. To address this, innovative strategies like Next-Generation Risk Assessment (NGRA) are being explored, integrating historical animal data with new mechanistic insights from non-animal New Approach Methodologies (NAMs). This paper introduces the TOXIN knowledge graph (TOXIN KG), a tool designed to retrieve toxicological information on cosmetic ingredients, with a focus on liver-related data.

A sustainable method for the synthesis of 2-deoxy lactones as direct precursors to 2-deoxy sugars via regioselective UV-light-driven dealkyloxylation of carbohydrate-derived lactones is detailed. This catalyst- and additive-free protocol utilizes light irradiation, providing high step economy and functional group compatibility. This environmentally friendly and straightforward approach enhances the synthetic toolbox for 2-deoxy sugars, which is vital in numerous biologically active molecules and drug candidates.

This work introduces PhysioEx, a Python library designed to support the analysis of sleep stages using deep learning and Explainable AI (XAI). Approach: PhysioEx provides an extensible and modular API for standardizing and automating the sleep staging pipeline, covering data preprocessing, model training, testing, fine-tuning, and explainability.

We present a visible-light-promoted radical cascade cyclization reaction sulfonylmethylation, cyano insertion, and radical cyclization of unactivated alkenes bearing cyano groups. This strategy enables the rapid synthesis of sulfonylmethylated phenanthridines under mild conditions with broad substrate compatibility, operational simplicity, and mild reaction conditions. The developed approach provides a novel pathway for assembling complex polycyclic nitrogen-containing frameworks, addressing a critical synthetic challenge and expanding the toolbox of photochemical transformations in organic synthesis.
