[Antitumor activity of polysaccharides extracted from two wild amanitas].

Objective: To investigate the anti-tumor activity of two wild Amanita strains, and to determine the inhibition ratio of their effective chemical components on tumor cells in vitro.

Methods: Two wild Amanita strains collected in Sichuan province were identified through morphology and molecular methods. The chemical components from the fruit body of the two strains were extracted with petroleum ether, chloroform, methanol and water. The anti-tumor activity of the extracts were tested on the cancer cells of MGC and Lovo strains. The polysaccharides were isolated with hot water and tested for its anti-tumor activity in vitro.

Results: The two wild Amanitas were identified as Amanita subjunquillea and Amanita pantherina. The water-soluble extracts from the Amanitas had anti-tumor effect, with polysaccharide as one of the main effective components. The IC50 value of the polysaccharide isolated from M1 was 3.790 mg/mL for the MGC cells.

Conclusion: Polysaccharides isolated from the two wild Amanitas have considerable anti-tumor activity against MGC cells in vitro.

SET domain bifurcated histone lysine methyltransferase 1 (SETDB1/ESET), a pivotal H3K9 methyltransferase, has been extensively studied since its discovery over two decades ago. SETDB1 plays critical roles in immune regulation, including B cell maturation, T-cell activity modulation, and endogenous retrovirus (ERV) silencing. Ultraviolet (UV)-induced DNA mutations produce genetic drivers of cutaneous melanoma initiation and numerous neoantigens that can trigger anti-tumor immune responses in the host. Consequently, melanoma cells must rapidly evolve to evade immune detection by simultaneously modulating cell-autonomous epigenetic mechanisms and tumor-microenvironment interactions. B7-H3 (CD276), a member of the B7-family of immune checkpoint proteins, has been shown to have immunological and non-immunological effects promoting tumorigenesis [1, 2] and expression correlates with poor prognosis for many solid tumors, including cervical, ovarian and breast cancers [3-6]. We recently identified a tumor-cell autochthonous tumorigenic role for dimerization of the 4Ig isoform of B7-H3 (4Ig-B7-H3) [7], where 4Ig-B7-H3 dimerization activated tumor-intrinsic cellular proliferation and tumorigenesis pathways, providing a novel opportunity for therapeutic intervention. Herein, a live cell split-luciferase complementation strategy was used to visualize 4Ig-B7-H3 homodimerization in a high-throughput small molecule screen (HTS) to identify modulators of this protein-protein interaction (PPI).
