N(α)-acetylation is a common protein modification catalyzed by different N-terminal acetyltransferases (NATs). Their essential role in the biogenesis and degradation of proteins is becoming increasingly evident. The NAT hNaa50p preferentially modifies peptides starting with methionine followed by a hydrophobic amino acid. hNaa50p also possesses N(ε)-autoacetylation activity. So far, no eukaryotic NAT has been mechanistically investigated. In this study, we used NMR spectroscopy, bisubstrate kinetic assays, and product inhibition experiments to demonstrate that hNaa50p utilizes an ordered Bi Bi reaction of the Theorell-Chance type. The NMR results, both the substrate binding study and the dynamic data, further indicate that the binding of acetyl-CoA induces a conformational change that is required for the peptide to bind to the active site. In support of an ordered Bi Bi reaction mechanism, addition of peptide in the absence of acetyl-CoA did not alter the structure of the protein. This model is further strengthened by the NMR results using a catalytically inactive hNaa50p mutant.

The CO oxidation reaction on (Co,Mg,Mn,Ni,Zn)(Al,Co,Cr,Fe,Mn)O and (Cr,Mn,Fe,Co,Ni)O high entropy spinel oxides was studied for what concerns its mechanism by means of soft X-ray absorption spectroscopy. In the (Cr,Mn,Fe,Co,Ni)O high entropy spinel, CO oxidation starts at 150 °C, and complete conversion to CO is obtained at 300 °C. For the (Co,Mg,Mn,Ni,Zn)(Al,Co,Cr,Fe,Mn)O spinel oxides, in contrast, the reaction starts at 200 °C, and complete conversion needs temperatures of the order of 350 °C.

In the past few years, the direct activation of organohalides by ligated boryl radicals has emerged as a potential synthetic tool for cross-coupling reactions. In most existing methods, ligated boryl radicals are accessed from NHC-boranes or amine-boranes. In this work, we report a new photocatalytic platform by modular assembly of readily available amines and diboron esters to access a library of ligated boryl radicals for reaction screening, thus enabling the cross-coupling of organohalides and alkenes including both activated and unactivated ones for C(sp)-C(sp) bond formation by using the assembly of DABCO A1 and BNepB1.
