A series of phenoxy linked bisquinoline derivatives were synthesised from the Friedlander annulation of 2-(4-acetylphenoxy)-1-aryl-1-ethanones with 2-aminobenzophenone in good yields using (±)-camphor-10-sulfonic acid (CSA) as the catalyst. These compounds were screened for their in vitro activity against Mycobacterium tuberculosis H37Rv (MTB) and among the 23 compounds screened, 2-(3-bromophenyl)-6-chloro-3-[4-(6-chloro-4-phenyl-2-quinolyl)phenoxy]-4-phenylquinoline (3q) and 2-(4-bromophenyl)-6-chloro-3-[4-(6-chloro-4-phenyl-2-quinolyl)phenoxy]-4-phenylquinoline (3o) were found to be the most active compounds with MIC of 1.1 and 2.2 μM against MTB. The cytotoxic effects against mouse fibroblasts (NIH 3T3) in vitro were evaluated for 3o and 3q, which displayed no toxic effects against mouse fibroblast cell line NIH 3T3.

Herein, we describe an air- and moisture-stable, homogeneous zinc catalyst stabilised using an electron deficient N^N^N pincer-type ligand. This ternary, penta-coordinated neutral molecular catalyst [Zn(N^N^N)Cl] selectively produces α-alkylated ketone derivatives (14 examples) through a one-pot acceptorless dehydrogenative coupling (ADC) reaction between secondary and primary alcohols using the borrowing hydrogen (BH) approach in good to excellent isolated yields (up to 93%). It is worth noting that this catalyst also provides an eco-friendly route for the synthesis of quinoline derivatives (30 examples) using 2-aminobenzyl alcohols as alkylating agents successive dehydrogenative coupling and N-annulation reactions.

An efficient, MnO-catalyzed ligand-free synthesis of ()-6,7-dihydrodibenzo[,][1,7]phenanthrolines, and ()-1,2,3,4-tetrahydrobenzo[][1,6]naphthyridines, utilizing, 2-amino-5-chloro-benzhydrol, acridinol, or 1-benzyl-4-piperidinol, , and benzyl alcohols, is reported. The MnO-catalyzed dehydrogenative Friedlander annulation utilizing ChCl/p-TSA (DES-1) and subsequent C(sp)-H functionalization with TBAB/p-TSA (DES-2) was effected at 100 °C. The optimized reaction conditions gave excellent product yields, and the products were evaluated for their by UV absorption and fluorescence emission studies.

The goal of this study was directed to synthesize a novel class of annulated compounds containing difuro[3,2-:3',2'-]chromene. Friedländer condensation of -aminoacetyl derivative was performed with some active methylene ketones, namely, 1,3-cyclohexanediones, pyrazolones, 1,3-thiazolidinones and barbituric acids, furnished furochromenofuroquinolines (,), furochromenofuropyrazolopyridines (-), furochromenofurothiazolopyridines (,) and furochromenofuropyridopyrimidines (, ), respectively. Also, condensation of substrate with 5-amine-3-methyl-1-pyrazole and 6-amino-1,3-dimethyluracil, as cyclic enamines, resulted in polyfused systems and , respectively.

Gold(III)-catalyzed annulation of electron-deficient alkynes and 2-amino-arylcarbonyls provides general modular one-step access to a broad scope of quinoline products. This highly selective reaction is a useful alternative to the classic Friedländer synthesis, which requires harsh reaction conditions. In contrast, the developed method works under relatively mild PicAuCl-catalyzed conditions and exhibits a high functional group tolerance (40 examples; yields of ≤96%).
