A group of novel synthetic indoloisoquinolines was prepared and its potential as a novel series of small-molecule anti-malarial leads was assessed. The structure-activity relationship on variation of three distinct regions of chemical space was investigated. A lead compound was generated with an activity close to that observed for a known anti-malarial natural product, dihydrousambarensine, that shares the indoloisoquinoline template structure.

The cholinergic deficits and amyloid beta (Aβ) aggregation are the mainstream simultaneously observed pathologies during the progression of Alzheimer's disease (AD). A series of isatin derivatives which could inhibit colorectal cancer (CRC) were synthesized. Among those compounds, 5B exhibited good inhibitory activity of CRC through the inhibition of tubulin expression, inducing apoptosis, and causing G2/M phase cell cycle arrest pathway, which suggested that 5B could be a potential tubulin inhibitor. Graph neural networks (GNNs) have shown great promise in modeling graph-structured data, but the over-smoothing problem restricts their effectiveness in deep layers. Two key weaknesses of existing research on deep GNN models are: (1) ignoring the beneficial aspects of intra-class smoothing while focusing solely on reducing inter-class smoothing, and (2) inefficient computation of residual weights that neglect the influence of neighboring nodes' distributions. To address these weaknesses, we propose a novel Smoothing Deceleration (SD) strategy to reduce the smoothing speed rate of nodes as information propagates between layers, thereby mitigating over-smoothing.
