We report a concise synthesis of A-factor, the prototypical γ-butyrolactone signalling compound of Streptomyces bacteria. In analogy to enzymatic reactions in A-factor biosynthesis, our synthesis features a tandem esterification-Knoevenagel condensation yielding a 2-acyl butenolide and a surprising, chemoselective conjugate reduction of this α,β-unsaturated carbonyl compound using sodium cyanoborohydride.

Doping and surface-modification are well-established strategies for the performance enhancement of bismuth vanadate (BiVO) photoanodes in photoelectrochemical (PEC) water splitting devices. Herein, a "double-use" strategy for the development of high-performance BiVO photoanodes for solar water splitting is reported, where a molecular cobalt-phosphotungstate (CoPOM = Na[Co(HO)(PWO)]) is used both as a bulk doping agent as well as a surface-deposited water oxidation cocatalyst. The use of CoPOM for bulk doping of BiVO is shown to enhance the electrical conductivity and improve the charge separation efficiency, resulting in the enhancement of the maximum applied-bias photoconversion efficiency (ABPE) by a factor of ∼18 to 0.

Balanced self-renewal and differentiation of stem cells are crucial for maintaining tissue homeostasis, but the underlying mechanisms of this process remain poorly understood. Here, from an RNA interference (RNAi) screen in adult Drosophila intestinal stem cells (ISCs), we identify a factor, Pax, which is orthologous to mammalian PXN, coordinates the proliferation and differentiation of ISCs during both normal homeostasis and injury-induced midgut regeneration in Drosophila. Loss of Pax promotes ISC proliferation while suppressing its differentiation into absorptive enterocytes (ECs).

Epithelial cells can become polyploid upon tissue injury, but mechanosensitive cues that trigger this state are poorly understood. Using an Madin Darby Canine Kidney (MDCK) cell knock-out/reconstitution system, we show that α-catenin mutants that alter force-sensitive binding to F-actin or middle (M)-domain promote cytokinesis failure and binucleation, particularly near epithelial wound-fronts. We identified Leucine Zipper Tumor Suppressor 2 (LZTS2), a factor previously implicated in abscission, as a conformation sensitive proximity partner of α-catenin.
