American ginseng is well known as a traditional herb and is commonly used in clinical decoctions. Owing to increasing demand for Panax notoginseng-based medicines and health products, establishing a fast, simple, and reliable assay to analyze the chemical differences between its root and rhizome is important. Although previous studies showed that the chemical and biological differences between the root and rhizome of P. notoginseng seem to be small, efforts should be taken to investigate such differences to ensure the safety and efficacy of the products.

Ginseng has been used for prevention and treatment of disease for thousands of years in China and many other Asian countries. Phytochemical studies have indicated that ginsenosides, polysaccharides, alkaloids, and phenolic acids are the active constituents of ginseng. Main and branch roots of ginseng exhibit distinct bioactive behavior.

Natural compounds with diverse structural skeletons have different ionization efficiencies and different mass spectrometry (MS) responses. Some key factors influencing the electrostatic field induced spray ionization (EFISI)-MS for a variety of natural compounds have been optimized and improved. Fifteen reference substances representing ten well-known skeletons of natural products including alkaloids, flavonoids, phenolic acids, lignans, coumarins, anthraquinones, monoterpenoids, sesquiterpenoids, diterpenoids and triterpenoids, were selected and investigated for their EFISI-MS responses on TLC plates using a dot-blot test.
