Developing cavity-based supramolecular hydrogels is in its infancy because not many such hydrogelators are available. Reported herein is our creation of rigid cavitand cyclotriveratrylene (CTV) based hydrogelators from the molecular backbones of CTVs that were in limited cases shown to form organogels. For doing so deprotonable -COOH or protonable -NH(2) was introduced as terminal group into the rigid and hydrophobic CTV backbones. We thus successfully obtained optically anisotropic supramolecular hydrogels from these new CTVs hydrogelators with excellent thermostability and high tolerance towards strong electrolytes. The obtained CTV-1 and CTV-2 hydrogels are luminescent and exhibit reversible gel-to-sol and sol-to-gel transitions upon pH variations. The success in creating CTV-1 and CTV-2 hydrogelators on the basis of the skeleton of a CTV-organogelator suggests that balancing the hydrophilic and hydrophobic characters of the ionic and hydrophobic moieties well in the gelator molecule is important for designing a promising hydrogelator.

Alkaline phosphatase (ALP) is a biomarker for many diseases, and monitoring its activity level is important for disease diagnosis and treatment. In this study, we used the microdroplet technology combined with an laser-induced polymerization method to prepare the Ag nanoparticle (AgNP) doped hydrogel microbeads (HMBs) with adjustable pore sizes that allow small molecules to enter while blocking large molecules. The AgNPs embedded in the hydrogel microspheres can provide SERS activity, improving the SERS signal of small molecules that diffuse to the AgNPs.

Saccharides and peptides with markedly disparate stereochemical features serve as pivotal chiral molecular partners in living systems. The importance of glycosylation in influencing glycopeptide self-assembly has been recognized. However, how different chiral combinations of saccharides and peptides influence the macroscopic hydrogel mechanics, fiber nanomechanics, asymmetric molecular packing, and thermodynamic changes during glycopeptide self-assembly remains unknown.

The efficacy of cancer immunotherapy relies on a sufficient amount of functional immune cells. Triple-negative breast cancer lacks enough immune cell infiltration, and adjuvant therapy is necessary to prime anti-tumor immunity. However, the improvement in efficacy is unsatisfactory with concern about inducing systemic immunotoxicity.

Persistent inflammation is a major cause of diabetic wounds that are difficult to heal. This is manifested in diabetic wounds with excessive reactive oxygen clusters (ROS), advanced glycation end products (AGE) and other inflammatory factors, and difficulty in polarizing macrophages toward inhibiting inflammation. Berberine is a natural plant molecule that inhibits inflammation; however, its low solubility limits its biological function through cytosis.

In this work, we investigate the pH-responsive behavior of multidomain peptide (MDP) hydrogels containing histidine. Small-angle X-ray scattering confirmed that MDP nanofibers sequester nonpolar residues into a hydrophobic core surrounded by a shell of hydrophilic residues. MDPs with histidine on the hydrophilic face formed nanofibers at all pH values tested, but the morphology of the fibers was influenced by the protonation state and the location of histidine in the MDP sequence.
