During Drosophila oogenesis the expression of the sulfotransferase Pipe in ventral follicle cells is crucial for dorsoventral axis formation. Pipe modifies proteins that are incorporated in the ventral eggshell and activate Toll signaling which in turn initiates embryonic dorsoventral patterning. Ventral pipe expression is the result of an oocyte-derived EGF signal which down-regulates pipe in dorsal follicle cells. The analysis of mutant follicle cell clones reveals that none of the transcription factors known to act downstream of EGF signaling in Drosophila is required or sufficient for pipe regulation. However, the pipe cis-regulatory region harbors a 31-bp element which is essential for pipe repression, and ovarian extracts contain a protein that binds this element. Thus, EGF signaling does not act by down-regulating an activator of pipe as previously suggested but rather by activating a repressor. Surprisingly, this repressor acts independent of the common co-repressors Groucho or CtBP.

The increasingly widespread application of next-generation sequencing (NGS) in clinical diagnostics and epidemiological research has generated a demand for robust, fast, automated, and user-friendly bioinformatics workflows. To guide the choice of tools for the assembly of full-length viral genomes from NGS datasets, we assessed the performance and applicability of four open-source bioinformatics pipelines (shiver-for which we created a user-friendly Dockerized version, referred to as dshiver; SmaltAlign; viral-ngs; and V-pipe) using both simulated and real-world HIV-1 paired-end short-read datasets and default settings. All four pipelines produced consensus genome assemblies with high quality metrics (genome fraction recovery, mismatch and indel rates, variant calling F1 scores) when the reference sequence used for assembly had high similarity to the analyzed sample.

A core dielectric cylindrical rod wrapped in a dielectric circular pipe whose outer surface is enclosed by a helical conducting strip grating that is skewed along the axial direction is herein analyzed using the asymptotic strip boundary conditions along with classical vector potential analysis. Targeted for use as a cylindrical holographic antenna, the resultant field solutions facilitate the aperture integration of the equivalent cylindrical surface currents to obtain the radiated far fields. As each rod section of a certain skew angle exhibits a distinct modal attribute; this topology allows for the distribution of the cylindrical surface impedance via the effective refractive index to be modulated, as in gradient-index (GRIN) materials.

The fatigue failure of a structure may occur under a multiaxial vibration environment; it is necessary to establish a better multiaxial fatigue life prediction model to predict the fatigue life of the structure. This study proposes a new model (MWYT) by introducing the maximum absolute shear stress into the WYT model. The feasibility of the MWYT model is verified by using the multiaxial fatigue experimental data of 304 stainless steel, Q235B steel, 7075-T651 aluminum alloy and S355J0 steel.

The petroleum industry is essential for supplying crude oil, which is vital for fuel and chemicals and drives substantial investments in technologies, especially in regard to increasing the durability of the drill strings used in wellbore construction. This study aims to establish and to validate a hardbanding technology for reconditioning NC50 tool joints subjected to wear, thereby increasing drill pipe durability and reducing the risk of failure during drilling, which can lead to ecological pollution, human safety issues, and financial costs. The hardbanding of the tool joints was carried out using the gas metal arc welding process (GMAW) with two different wear-resistant wires, ARNCO 100XT and FLUXOFIL M58.
