Feline immunodeficiency virus (FIV)-based lentiviral vectors are useful for introducing integrated transgenes into nondividing human cells. This protocol describes the production of FIV-based lentiviral vectors using cells grown in CF10 or CF2 devices. It also details the harvesting and concentration of these vectors. The methods are for production for basic science laboratory use and in vivo experimentation. They do not meet standards for clinical-grade (good manufacturing practice [GMP]) production.

In cats, mycobacteriosis tends to present in a syndromic manner, with cases either being due to tuberculosis (TB) (in countries where TB is endemic), one of the "leprosy-like" diseases affecting the skin and subcutis, panniculitis caused by infection of subcutaneous tissues generally with rapidly growing Mycobacteria spp. or widely disseminated granulomatous disease, which is usually caused by members of the Mycobacterium avium-intracellulare complex (MAC). Disseminated MAC disease is rare, but when it occurs, usually develops in immunocompromised hosts with defective cell-mediated immunity.

Background: Feline immunodeficiency virus (FIV) is a highly mutable retrovirus with numerous species-specific strains, such as those infecting domestic cats (Felis catus, FIV) and wild populations of nondomestic cats including lions (Panthera leo, FIV). When FIV infection manifests as disease in domestic cats, common presentations include lymphadenopathy, lymphoid malignancies, oral cavity disease, immunological dysfunctions and neurological abnormalities. In contrast to domestic cats, there has been little published evidence of direct FIV disease associations in lions.
