Nitric oxide scavenging modulates mitochondrial dysfunction induced by hypoxia/reoxygenation.

The objective of the present study was to delineate the role of excessive accumulation of mitochondrial nitrogen species contributing to oxidative stress induced by hypoxia/reoxygenation in isolated mitochondria. The present study shows that incubation of isolated rat heart mitochondria under hypoxic, but not anoxic conditions, followed by reoxygenation decreases the rate of mitochondrial oxygen consumption, mitochondrial membrane potential, and calcium retention capacity. These alterations were prevented, at least in part, by 2-(4-carboxyphenyl)-4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide (carboxy-PTIO), a nitric oxide (NO) scavenger, N(G)-nitro-L-arginine-methyl ester (L-NAME), a broad-spectrum NO synthase inhibitor, or tempol, a superoxide dismutase mimetic and catalytic scavenger of peroxynitrite-derived radicals. In conclusion, these findings suggest a crucial role for nitric oxide pathways in cardiac oxidative stress induced by hypoxia/reoxygenation.

We previously reported the significant increase in limb muscle strength and cross-sectional area of the type IIb muscle fibers in the extensor digitorum longus (EDL) muscle in a type 2 diabetic animal model, with Spontaneously Diabetic Torii (SDT) fatty rats (n = 6) undergoing regular treadmill exercise from 8 to 16 weeks of age compared with sedentary SDT fatty rats (n = 6). This study investigated the mechanism by which exercise training prevented skeletal muscle wasting in the EDL muscle of the SDT fatty rats. The endurance exercise for 8 weeks downregulated the expression of muscle RING-finger protein-1 (an E3 ubiquitin ligase) and upregulated the expression of CD31, insulin receptor substrate-2, and phosphorylated endothelial nitric oxide synthase in the EDL muscle of 16-week-old SDT fatty rats.

Calcium-based nanomaterials-mediated Ca overload-induced pyroptosis and its application in tumor therapy have received considerable attention. However, the calcium buffering capacity of tumor cells can maintain mitochondrial calcium homeostasis, so it is important to effectively disrupt this homeostasis to activate pyroptosis. Here, a nano-modulator CUR@CaCO-PArg@HA (CCAH) was developed to regulate calcium overload in multiple channels and activate pyroptosis.

This study comprehensively explores the relationship between the structure of carboxymethyl-pachymaran (CMP) and its diverse biological activities, including immunomodulation, anti-inflammatory effects, tumor cell proliferation inhibition, and antioxidant activity. By adjusting preparation parameters, highly purified CMP samples with varying degrees of substitution (DS) and molecular weights (Mw) were successfully obtained. The results indicate that CMP, composed primarily of β-D-glucan, exhibits different levels of activity depending on its structural characteristics.
