The purpose of this work was to study the dynamics of expression of mitochondrial Mn-dependent superoxide dismutase (Mn-SOD) 3 and 24 hours after single and triple exposure to mild hypoxia. The investigation was conducted in 18 male Wistar rats using immunocytochemical method. It was shown that in various hippocampal areas the effects of single and triple hypoxia exposure on the Mn-SOD expression could be different or largely similar. The expression dynamics at four time points studied (3 and 24 hours after the first exposure, 3 and 24 hours after the third one) had a wave character which may be important for the development of moderate hypoxia-induced tolerance to subsequent more severe exposures.

Background: The combination of conventional chemotherapy and immune checkpoint inhibitors (ICIs) has been unsuccessful for pancreatic ductal adenocarcinoma (PDAC). Administration of maximum tolerated dose of chemotherapy drugs may have immunosuppressive effects.
Methods: We thus tested, by using the preclinical model of PDACs including the genetically engineered mouse KPC spontaneous pancreatic tumor model and the pancreatic KPC tumor orthotopic implant model, the combinations of synthetic innate immune agonists including STING and NLRP3 agonist, respectively, and ICIs with or without chemotherapy.

Immunity suffers a function deficit during aging, and the incidence of cancer is increased in the elderly. However, most cancer models employ young mice, which are poorly representative of adult cancer patients. We have previously reported that Triple-Therapy (TT), involving antigen-presenting-cell activation by vinorelbine and generation of TCF1-stem-cell-like T cells (scTs) by cyclophosphamide significantly improved anti-PD-1 efficacy in anti-PD1-resistant models like Triple-Negative Breast Cancer (TNBC) and Non-Hodgkin's Lymphoma (NHL), due to T-cell-mediated tumor killing.
