Xpert MTB/RIF-generated cycle-threshold (C(T)) values have poor clinical utility as a rule-in test for smear positivity (cut-point ≤20.2; sensitivity 32.3%, specificity 97.1%) but moderately good rule-out value (cut-point >31.8; negative predictive value 80.0%). Thus, 20% of individuals with C(T) values >31.8 were erroneously ruled out as smear-negative. This group had a significantly lower sputum bacillary load relative to correctly classified smear-positive patients (C(T) ≤ 31.8; P < .001). These data inform on public health and contact tracing strategies.

Purpose: Using a fully automated multitask deep learning method, which enabled simultaneous segmentation and quantification of all major anterior segment structures with swept-source optical coherence tomography (SS-OCT), we aimed to investigate the three-dimensional (3D) alterations in iris morphology before and after implantable collamer lens (ICL) surgery.
Methods: All enrolled patients underwent anterior segment SS-OCT (ANTERION) within one week before and after ICL surgery. A multitask network automatically performed iris SS-OCT image segmentation and quantitative measurements of 3D iris morphology (iris thickness and volume of the inner 1-mm annular area and the outer 1-2-mm annular area, iris curvature [I-Curve], and iris smooth index [SI]).

Imaging the entire cardiomyocyte network in entire small animal hearts at single cell resolution is a formidable challenge. Optical microscopy provides sufficient contrast and resolution in 2d, however fails to deliver non-destructive 3d reconstructions with isotropic resolution. It requires several invasive preparation steps, which introduce structural artefacts, namely dehydration, physical slicing and staining, or for the case of light sheet microscopy also clearing of the tissue.

Mitochondria are double membrane-bound organelles essential for generating energy in eukaryotic cells. Mitochondria can be readily visualized in 3D using Volume Electron Microscopy (vEM), and accurate image segmentation is vital for quantitative analysis of mitochondrial morphology and function. To address the challenge of segmenting small mitochondrial compartments in vEM images, we propose an automated mitochondrial segmentation method called GCTransNet.
