DNA-binding and photocleavage studies of ruthenium(II) complexes containing asymmetric intercalative ligand.

A novel asymmetric ligand 2-(pyridine-2-yl)-1-H-imidazo[4,5-i]dibenzo[2,3-a:2',3'-c]phenazine (pidbp) and its ruthenium complexes [Ru(L)(2)(pidbp)](2+) (L=bpy (2, 2'- bipyridine), phen (1, 10 - phenanthroline)), have been synthesized and characterized by elemental analysis, ES-MS, (1)H NMR. Various methods support the conclusion that both Ru(II) complexes can intercalate into DNA base pairs. Complex [Ru(bpy)(2)(pidbp)](2+)4 exhibits its DNA "molecular light switch" properties. Furthermore, the two complexes are efficient DNA-photocleavers under irradiation at 365 nm, and complex 5 exhibits a stronger DNA-photocleavage efficiency than complex 4. The mechanism of DNA cleavage is an oxidative process by generating singlet oxygen.

Two Ru(II) complexes, [Ru(pydppn)(bim)(py)] [; pydppn = 3-(pyrid-2'-yl)-4,5,9,16-tetraaza-dibenzo[]naphthacene; bim = 2,2'-bisimidazole; py = pyridine] and [Ru(pydppn)(Mebim)(py)] [; Mebim = 2,2'-bis(4,5-dimethylimidazole)], were synthesized and characterized, and their photophysical properties, DNA binding, and photocleavage were evaluated and compared to [Ru(pydppn)(bpy)(py)] (; bpy = 2,2'-bipyridine). Complexes and exhibit broad MLCT (metal-to-ligand charge transfer) transitions with maxima at ∼470 nm and shoulders at ∼525 and ∼600 nm that extend to ∼800 nm. These bands are red-shifted relative to those of , attributed to the π-donating ability of the bim and Mebim ligands.

A set of arylazo sulfones, known to undergo N-S bond cleavage upon light exposure, has been synthesized, and their activity in the dark and upon irradiation towards DNA has been investigated. Their interaction with calf-thymus DNA has been examined, and the significant affinity observed (most probably due to DNA intercalation) was analyzed by means of molecular docking "in silico" calculations that pointed out polar contacts, mainly via the sulfonyl moiety. Incubation with plasmid pBluescript KS II revealed DNA cleavage that has been studied over time and concentration.

Two novel halogenated (Br- and F-) quinazoline derivatives, namely [()-4-(2-((6-bromopyridin-2-yl)methylene)hydrazinyl)quinazoline] (L1) and [()-4-(2-((3-fluoropyridin-2-yl)methylene)hydrazinyl) quinazoline] (L2), were synthesized and characterized. Their interaction with a series of metal(II) ions (= Mn(II), Ni(II), Cu(II), Zn(II) and Cd(II)) resulted in the formation of six mononuclear complexes characterized by spectroscopic techniques and single-crystal X-ray crystallography. The complexes bear the formulae [Ni(L)](NO) (1), [Zn(L)](NO)(PF) (2), [Cd(L)(HO)(CHOH)(NO)](NO) (3), [Cu(L)Cl] (4), [Ni(L)](NO) (5) and [Mn(L)(CHOH)(Cl)] (6).
