Natural plant compounds have an unexceptional influence in pharmacy as they provide an uncountable number of invaluable lead molecules. Phytochemical researches nowadays focus on bio-assay guided revealing of the therapeutic profile and synergism of medicinal herbs and their constituents. Assessing the clinical and biological potential and determining the pharmacokinetics of herbal constituents is also an area of much interest. This work was conducted in order to carry out a sensitive liquid chromatography tandem mass spectrum (HPLC-MS/MS) method for the pharmacokinetics study of (+)-pinoresinol-di-β-D-glucopyranoside (PG) in rats' plasma after oral administration of Eucommia ulmoides Oliv extract. The validated method was by means of linearity, precision, matrix effect and recovery so that it could be used for the pharmacokinetic study of PG. The obtained pharmacokinetic parameters shown that PG pertains to one-compartment model and 95% of PG was eliminated within 12h.

The exploration of the mitochondrial apoptotic pathway in living cells is of great significance for achieving tumor diagnosis and treatment. However, visualization of the mitochondrial apoptotic pathway induced by specific proteins has rarely been reported. In this paper, we designed and synthesized a fluorescent probe Cy-JQ1 based on the bromodomain-containing protein 4 (BRD4) inhibition.

Background: ATR is an apical DDR kinase activated at damaged replication forks. Elimusertib is an oral ATR inhibitor and potentiates irinotecan in human colorectal cancer models.

Methods: To establish dose and tolerability of elimusertib with FOLFIRI, a Bayesian Optimal Interval trial design was pursued.

Aims: The beta-lactam antibiotic temocillin is increasingly used to treat extended-spectrum beta-lactamase (ESBL-producing) strains; however, its protein binding is complex. This study aims to predict unbound temocillin concentrations in various participant groups to determine its impact on the probability of target attainment (PTA) and to improve dosing recommendations.

Methods: The plasma pharmacokinetics were analysed using non-linear mixed-effects modelling.

Objective: Previous experiments have demonstrated that BGM0504, a GLP-1R/GIPR dual agonist drug by molecular dynamics-guided optimization, had enhanced agonistic activity compared to tirzepatide. This study aims to investigate its safety, tolerability, pharmacokinetics (PK) and pharmacodynamics (PD) in Chinese healthy volunteers.

Methods: A randomized, double-blind, placebo-controlled and dose-escalation Phase I study was conducted as follows: a single dose (2.

Pharmacogenetic and pharmacokinetic factors for dexmedetomidine-associated hemodynamic instability in pediatric patients.

Front Pharmacol

January 2025

Institute of Clinical Pharmacology, School of Pharmaceutical Sciences, Sun Yat-Sen University, Guangzhou, Guangdong Province, China.

Purpose: The incidence of hemodynamic instability associated with dexmedetomidine (DEX) sedation has been reported to exceed 50%, with substantial inter-individual variability in response. Genetic factors have been suggested to contribute significantly to such variation. The aim of this study was to identify the clinical, pharmacokinetic, and genetic factors associated with DEX-induced hemodynamic instability in pediatric anesthesia patients.
