Context/objective: To report an association between epithelial growth factor receptor (EGFR) inhibitors and ocular side effects.
Materials And Methods: Collection of spontaneous reports at the National Registry of Drug-Induced Ocular Side Effects (Casey Eye Institute, Portland, Oregon) in conjunction with a literature review of EGFR inhibitors associated with ocular adverse drug reactions (ADR).
Results: EGFR inhibitors are associated with conjunctivitis, meibomitis, dry eyes, periocular skin changes and trichomegaly. EGFR inhibitors may also cause superficial punctate corneal changes and corneal erosions.
Discussion: This is the first overview of all known ocular side effects associated with the use of marketed EGFR inhibitors. This is also the first effort for this class of drugs using the World Health Organization Classification as to causality.
Conclusion: Ophthalmologists should be aware of possible adverse ocular side effects from EGFR inhibitors and treat based on the type of ADR encountered. All ocular side effects noted are fully reversible upon discontinuation of the drug.

Recurrent missense mutations in the human epidermal growth factor receptor 2 (HER2) have been identified across various human cancers. Among these mutations, the active S310F mutation in the HER2 extracellular domain stands out as not only oncogenic but also confers resistance to pertuzumab, an antibody drug widely used in clinical cancer therapy, by impeding its binding. In this study, we have successfully employed computational-aided rational design to undertake directed evolution of pertuzumab, resulting in the creation of an evolved pertuzumab variant named Ptz-SA.

Objective: This study focuses on epidermal growth factor receptor-mutated lung adenocarcinoma, known for frequent brain metastasis. It aimed to compare the clinical outcomes and cost-effectiveness of combining Gamma Knife radiosurgery (GKRS) with tyrosine kinase inhibitors (TKIs) (GKRS+TKI group) versus TKIs alone (TKI group) for the treatment of patients with newly diagnosed brain metastasis in this condition.
Methods: Study characteristics of the two groups were matched using inverse probability of treatment weighting (IPTW).

Based on network pharmacology and molecular docking methods, this study explored its active compounds and confirmed its potential mechanism of action against Hand-foot skin reaction induced by tumor-targeted drugs. Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform and UniProt Database were used to obtain the active ingredients and target proteins of Spatholobi Caulis. All hand-foot skin reaction (HFSR)-related targets were obtained with the help of the Human Gene Database, Online Mendelian Inheritance in Humans (OMIM), DisGeNET and DrugBank databases.

Osimertinib, a third-generation epidermal growth factor receptor (EGFR)-tyrosine kinase inhibitor (TKI), has revolutionized one of the standard most efficient treatments for EGFR mutation-positive non-small cell lung cancer (NSCLC). Osimertinib, a third-generation EGFR-TKI, is currently one of most efficient treatments in clinical practice. However, it has a potentially fatal side effect: interstitial lung disease (ILD).
