Flavonoids: A versatile source of anticancer drugs.

An exponential increase in the number of studies investigating how different components of the diet interact at the molecular and cellular level to determine the fate of a cell has been witnessed. In search for anticancer drugs compelling data from laboratories, epidemiologic investigations, and human clinical trials showed that flavonoids have important effects on cancer chemoprevention and chemotherapy. In many molecular mechanisms of action for prevention against cancer, flavonoids play a major role by interacting between different types of genes and enzymes. Many mechanisms of action have been identified, including carcinogen inactivation, antiproliferation, cell cycle arrest, induction of apoptosis, inhibition of angiogenesis, antioxidation, and reversal of multidrug resistance or a combination of these mechanisms. This review focuses on the anticancer activity of flavonoids as well as their molecular mechanisms, including the treatment of mammary and prostate cancer. This review also highlights some advanced derivatives of flavonoids, which play an important role against cancer.

Fullerenes (C, C) as carbon nanomaterials can enter the environment through natural processes and anthropogenic activities, while synthetic fullerenes are commonly used in medicine in targeted therapies in association with antibodies, or anticancer and antimicrobial drugs. As the nanoparticles, they can pass through cell membranes and organelles and accumulate in the entire cytoplasm. The red-fluorescent, water-soluble [70]fullerene derivative C-OMe-ser, which produces reactive oxygen species upon illumination with an appropriate wavelength, passed into the cytoplasm of the middle region in the digestive system.

Although there has been significant progress in current comprehensive anticancer treatments centered on surgery, postoperative recurrence and tumor metastasis still significantly affect both prognosis and quality of life of the patient. Hence, the development of precisely targeted tumor therapies and exploration of immunotherapy represent ideal strategies for tumor treatment. Photodynamic therapy (PDT) is a localized and relatively safe treatment modality that not only induces multiple modes of tumor cell death but also mediates the secondary immunological responses against tumor resistance and metastasis.

Quinolines have emerged as essential components in various medicinal agents, playing a key role in treating various ailments. Numerous drugs with a quinoline core have been recognized for their antimalarial, antibacterial, and anticancer activities and have been successfully commercialized, including chloroquine, ciprofloxacin, topotecan, etc. Over the past two decades, we have witnessed a tremendous expansion in the C-H bond functionalization of quinoline scaffolds to widen this chemical space for drug discovery further.

The blood-brain barrier (BBB) presents one of the main obstacles to delivering anticancer drugs in glioblastoma. Herein, we investigated the potential of a series of cyclic ruthenium-peptide conjugates as photoactivated therapy candidates for the treatment of this aggressive tumor. The three compounds studied, , , and ([Ru(Phphen) Ac-XRGDX-NH)]Cl with Phphen = 4,7-diphenyl-1,10-phenanthroline and X, X = His or Met), include an integrin-targeted pentapeptide coordinated to a ruthenium warhead via two photoactivated ruthenium-X bonds.
