Aims: To determine efficacy of the analgesic flupirtine in the treatment of overactive bladder syndrome in a proof-of-concept study.
Methods: Double-blind, double-dummy, three-armed comparison of flupirtine extended release (400 mg/day, titrated to 600 mg/day), tolterodine extended release (4 mg/day) and placebo for 12 weeks.
Results: When major elevations of liver enzymes (more than three times the upper normal limit) were detected in several flupirtine-exposed patients, the study was prematurely discontinued. Based on study-end data, hepatotoxicity was detected in 31% of patients receiving flupirtine for ≥ 6 weeks.
Conclusions: Unexpected frequent and relevant toxicity can occur when testing an established drug for a new indication.

Background: Data on the occurrence of unexpected sudden cardiac death (SCD) in different seasons are limited.
Methods: All unexpected sudden death victims have to undergo medico-legal autopsy obligated by the Finnish law. Consecutive series of all unexpected autopsied SCD victims (n=5,869) were prospectively collected from the geographically defined area in the Northern Finland during a twenty years period from 1998 to 2017.

Ten-eleven translocation (TET) enzymes oxidize 5-methylcytosine (mC) in DNA, contributing to the regulation of gene transcription. Diverse mutations of TET2 are frequently found in various blood cancers, yet the full scope of their functional consequences has been unexplored. Here, we report that a subset of TET2 mutations identified in leukemia patients alter the substrate specificity of TET2 from acting on mC to thymine.

This paper reports a theoretically-driven quantitative content analysis of news media discourse on climate change, its effects, and solutions to understand how US news discourse differs from widely supported scientific conclusions on global climate. Despite the dire warnings and calls to action, US public opinion on the causes and solutions to climate change remain divided. In the global context, the US's split views are anomalous and may be an artifact of the US media's coverage of the climate crisis.
