The X-ray crystal structure, NMR binding studies, and enzyme activity of silver(I) metallated hen egg white lysozyme are presented. Primary bonding of silver is observed through His15 with secondary bonding interactions coming from nearby Arg14 and Asp87. A covalently bound nitrate completes a four coordinate binding pocket.

The decline in eggshell quality with increasing hen age may be related to changes in ultrastructure and chemical composition, with matrix proteins playing key roles in these changes. Hen egg-white lysozyme (HEWL) is commonly used in food preservation and as a substitute for sulfur dioxide in wine production and to prevent late blowing defects in cheese production. However, HEWL is an egg allergen that can cause severe allergic reactions in allergic individuals after accidental ingestion. Therefore, in order to prevent life-threatening health problems caused by consumer allergies, we developed an extremely specific gold immunochromatographic assay (GICA) tolerant to matrix effects for the qualification and quantification of HEWL in food products.

Purpose: This study examines the interaction between benzoylmesaconine (BMA) and hen egg white lysozyme (HEWL) under various physiological conditions, aiming to determine how BMA affects the HEWL's structure and function.
Methods: Several analytical techniques were used, including tryptophan assay, light scattering, thioflavin T (ThT)-binding assay, dynamic light scattering, 8-anilino-1-naphthalenesulfonic acid (ANS)-binding assay, circular dichroism (CD) spectroscopy, enzyme activity assay, and molecular docking.
Results: The tryptophan assay displayed a concentration-dependent decrease in tryptophan fluorescence, showing an interaction between BMA and HEWL.
