BnRCH gene inhibits cell growth of Hela cells through increasing the G2 phase of cell cycle.

The ubiquitin-proteasome pathway (UPP) is an important protein degradation system universally existing in eukaryotic organisms from yeast to human. In this system, hundreds of E3 ubiquitin-protein ligases are most important because they provide the substrate specificity and control many cellular processes. UPP has been found to be relevant to cancer development. BnRCH, the protein product from a novel gene isolated from Brassica napus, also has E3 ubiquitin-protein ligase activity. In order to exploit its potential use, human cervical carcinoma cell Hela (Hela cells) was transiently and stably transfected with BnRCH. The experimental results demonstrated: (1) in Hela cells, BnRCH inhibited the cell growth of Hela cells and increased their sensitivity to the anti-cancer chemotherapeutic drug cisplatin; and (2) the growth inhibition effect of BnRCH in Hela cells was found due to G2 phase cell cycle arrest with the transcriptional up-regulation of p21 (waf1/cip1), rather than apoptosis. This research suggests BnRCH has potential use in cancer therapy.

Introduction: Several aspects of the involvement of HPV in the pathogenesis of HPV-associated diseases remain poorly understood including mechanistic aspects of infection and the question of why the majority of HPV-positive HNSCC-patients are non-smokers, whereas HPV-negatives are smokers. Our previous research, based on 1,100 patient samples, hypothesized an explanation for this phenomenon: Smoking induces upregulation of a mucosal protective protein (SLPI), which competes with HPV for binding to Annexin A2 (AnxA2), pivotal for HPV cell entry. Novel thiazole analogs 3a, 3b, 4, 5, 6a-g, 8a, 8b, 9a-c, 10a-d and 11 were designed and synthesized as molecular mimetics of sunitinib. antitumor activity of the obtained compounds was investigated against HepG2, HCT-116, MCF-7, HeP-2 and HeLa cancer cell lines. The obtained data showed that compounds 3b and 10c are the most potent members toward HepG2, HCT-116, MCF-7 and HeLa cells.

Centromeres are marked by the centromere-specific histone H3 variant CENP-A/CENH3. Throughout the cell cycle, the constitutive centromere-associated network is bound to CENP-A chromatin, but how this protein network modifies CENP-A nucleosome conformations in vivo is unknown. Here, we purify endogenous centromeric chromatin associated with the CENP-C complex across the cell cycle and analyze the structures by single-molecule imaging and biochemical assays.

A series of new Pd(II) complexes were synthesized from the reaction of andrographolide appended hydrazide derivatives with potassium tetrachloropalladate K[PdCl]. The formation of the complexes was confirmed through structural assessments conducted using various spectroscopic techniques. From the spectral studies we confirmed that the ligands coordinated to Pd(II) ion via amine nitrogen and enone oxygen.
