Background: Since spermatogonial stem cell transplantation (SSCT) and testicular tissue grafting (TTG) may have important clinical applications, the safety of these promising techniques has to be proved. This study was designed to characterize epigenetic modifications in prepubertal and adult mouse germ cells and to study these epigenetic mechanisms after SSCT and TTG.
Methods: Testicular cell suspensions were transplanted to the testes of genetically sterile W/W(v) recipients. Intratesticular tissue grafting was performed between green fluorescent protein (GFP(+)) donors and GFP(-) acceptor mice. DNMT1 and DNMT3A expression, the general methylation status and the histone modifications H3K4me3, H3K9ac, H4K5ac, H4K8ac, H4K12ac and H4K16ac were studied in a stage-dependent manner by immunohistochemistry and compared with data from adult control mice.
Results: The expression levels of DNMT1 and DNMT3A, the DNA methylation status and most of the stage-specific histone modifications after SSCT or TTG were not different from fertile adult controls. Although, in elongated spermatids, the acetylation pattern was as expected, the stage-dependent expression of H4K5ac and H4K8ac was altered after SSCT.
Conclusions: Intratesticular tissue grafting might be the better choice for fertility restoration. Disrupting the stem cell niche might influence epigenetic patterns. Since the function of H4K5ac and H4K8ac in spermatogonia and spermatocytes still has to be explored, in-depth epigenetical analyses are warranted.

Purpose: The aim of the present study is to examine the demographic data and clinical features of ocular surface injuries due to thermal burns and to evaluate LSCD in the light of global consensus.
Methods: Thirty-three eyes of 20 cases with ocular surface injury due to thermal burn who attended to the clinic between 2012 and 2023 were included in the study. LSCD severity was staged according to the global consensus which was published in 2019.

Autoimmune liver diseases (AILD) involve dysregulated CD4 T cell responses against liver self-antigens, but how these autoreactive T cells relate to liver tissue pathology remains unclear. Here we perform single-cell transcriptomic and T cell receptor analyses of circulating, self-antigen-specific CD4 T cells from patients with AILD and identify a subset of liver-autoreactive CD4 T cells with a distinct B-helper transcriptional profile characterized by PD-1, TIGIT and HLA-DR expression. These cells share clonal relationships with expanded intrahepatic T cells and exhibit transcriptional signatures overlapping with tissue-resident T cells in chronically inflamed environments.

Frostbite injury refers to cold tissue injury which typically affects the peripheral areas of the body, and is associated with limb loss and high rates of morbidity. Historically, treatment options have been limited to supportive care, leading to suboptimal outcomes for affected patients. The pathophysiology of frostbite injury has been understood in recent years to share similarity with that of cold ischemia-reperfusion injury as seen in solid organ transplantation, of which mitochondria play an important contributing role.

Myelomeningocele (MMC) is a congenital defect of the spine characterized by meningeal and spinal cord protrusion through open vertebral archs, and its exposure to the amniotic fluid. Given that the progression of neuronal loss begins early in fetal life, an early coverage of the defect is required to improve the neurological outcomes. Several studies have proposed patches as an alternative to full surgical repair, to achieve an early protection of the spine and possibly reduce the rate of complications of current prenatal surgical procedures.
