Tryptophan-independent mutations were induced in CM1141 trpE65 umuC122::Tn5 following exposure to ultraviolet light (UV) plus delayed photoreversal. The mutations appeared to be exclusively class 2 ochre suppressors and showed mutation frequency decline (MFD) when the bacteria were incubated in glucose-salts medium after UV and before photoreversal. The phenomenon was similar to MFD after normal UV mutagenesis of umu+ bacteria, being inhibited in the presence of caffeine or in the absence of glucose.

The effect of a previously isolated antisuppressor mutation from bakers' yeast, that reduced the efficiency of the tyrosine-inserting ochre suppressor, SUP7-o, on other tyrosine-inserting ochre suppressors has been determined. As expected, the antisuppressor mutation, mod5-1, restricted the capacity of all eight tyrosine-inserting ochre suppressors to suppress nonsense mutations. Based on the suppression of five ochre alleles in the presence of mod5, the eight class I suppressors can be grouped into three subclasses.
View Article and Find Full Text PDFWe have studied the reversion of 8 nonsense alleles located in 7 different genes of Schizosaccharomyces pombe using 4-nitroquinoline-1-oxide (NQO) as a mutagenic agent. The nonsense mutants of S. pombe have been classified according to their suppressibility by defined opal and ochre suppressors into a class of efficiently suppressed opal and a class of inefficiency suppressed ochre mutants.
View Article and Find Full Text PDFSome preparations of beta-galactosidase from strains of Escherichia coli carrying point mutations in their lacZ genes did not precipitate with antibody as effectively as wild-type enzyme, but did not appear to be chain-terminating mutations as judged by polarity measurements and suppression. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis of crude extracts of induced Lac+ strains revealed that the monomer of beta-galactosidase ran as a band uncontaminated by other cellular proteins. This method was used to identify missense mutations in the alpha and beta portions of the lacZ gene.
