Cortico-thalamo-cortical circuits mediate sensation and generate neural network oscillations associated with slow-wave sleep and various epilepsies. Cortical input to sensory thalamus is thought to mainly evoke feed-forward synaptic inhibition of thalamocortical (TC) cells via reticular thalamic nucleus (nRT) neurons, especially during oscillations. This relies on a stronger synaptic strength in the cortico-nRT pathway than in the cortico-TC pathway, allowing the feed-forward inhibition of TC cells to overcome direct cortico-TC excitation. We found a systemic and specific reduction in strength in GluA4-deficient (Gria4(-/-)) mice of one excitatory synapse of the rhythmogenic cortico-thalamo-cortical system, the cortico-nRT projection, and observed that the oscillations could still be initiated by cortical inputs via the cortico-TC-nRT-TC pathway. These results reveal a previously unknown mode of cortico-thalamo-cortical transmission, bypassing direct cortico-nRT excitation, and describe a mechanism for pathological oscillation generation. This mode could be active under other circumstances, representing a previously unknown channel of cortico-thalamo-cortical information processing.

The output of the basal ganglia to the corticothalamic system plays an important role in regulating absence seizures. Inspired by experiments, we systematically study the crucial roles of two newly identified direct inhibitory striatal-cortical projections that project from the striatal D1 nucleus (SD1) and striatal D2 nucleus (SD2) to the cerebral cortex, in controlling absence seizures. Through computational simulation, we observe that typical 2-4 Hz spike and wave discharges (SWDs) can be induced through the pathological mechanism of cortical circuits, and both enhancing the inhibitory coupling weight on the striatal-cortical projections and improving the discharge activation level of striatal populations can effectively control typical SWDs.

A compact description of the frequency structure and topography of human alpha-band rhythms is obtained by use of the first four brain activity eigenmodes previously derived from corticothalamic neural field theory. Just two eigenmodes that overlap in frequency are found to reproduce the observed topography of the classical alpha rhythm for subjects with a single, occipitally concentrated alpha peak in their electroencephalograms. Alpha frequency splitting and relative amplitudes of double alpha peaks are explored analytically and numerically within this four-mode framework using eigenfunction expansion and perturbation methods.

Predictive processing theories conceptualize neocortical feedback as conveying expectations and contextual attention signals derived from internal cortical models, playing an essential role in the perception and interpretation of sensory information. However, few predictive processing frameworks outline concrete mechanistic roles for the corticothalamic (CT) feedback from layer 6 (L6), despite the fact that the number of CT axons is an order of magnitude greater than that of feedforward thalamocortical (TC) axons. Here we review the functional architecture of CT circuits and propose a mechanism through which L6 could regulate thalamic firing modes (burst, tonic) to detect unexpected inputs.

Neurons in the dorsolateral geniculate nucleus (dLGN) of the thalamus receive a substantial proportion of modulatory inputs from corticothalamic (CT) feedback and brain stem nuclei. Hypothesizing that these modulatory influences might be differentially engaged depending on the visual stimulus and behavioral state, we performed in vivo extracellular recordings from mouse dLGN while optogenetically suppressing CT feedback and monitoring behavioral state by locomotion and pupil dilation. For naturalistic movie clips, we found CT feedback to consistently increase dLGN response gain and promote tonic firing.
