Objective: To explore material basis of the pharmacological differences between the roasting and pro-roasting Raphani Semen.

Method: The two new sulfur-containing compounds (A209 and B221) were found changed after processing in the water decoction. The common precursor-C3 of A209 and B211 and the precursor of C3 were seperated and purified. Their transforming relationship was proved.

Result: The result showed that glucosinolates could decompose into sulforaphane and transform into A209 and B221 further in the boiling process.

Conclusion: This study provides some experimental evidences for revealing the mechanism of Raphani Semen processing.

Background: The presence of distant metastasis at the time of initial diagnosis is a prevalent issue in non-small cell lung cancer (NSCLC), affecting around 30-40 % of the patients. Acidic tumor microenvironment (TME) provides favorable conditions that increase the invasiveness and aggressiveness of NSCLC. The activity of the glycolytic enzyme lactate dehydrogenase (LDHA) increases intracellular lactate accumulation, which creates an acidic TME.

RSA-1 is a polysaccharide obtained from Raphani semen with a relatively clear structure and anti-colon cancer activity. In this study, high-performance liquid chromatography (HPLC), gas chromatography-mass spectrometry (GC-MS), and nuclear magnetic resonance (NMR) spectroscopy were applied to characterise the complex chain structure of RSA-1. Subsequently, the inhibitory effect on colon cancer growth through apoptosis induction in colon cancer cells was explored.

A new compound named raphanised A (), along with two known methylsulfinyl -butanyl derivatives () and seven known indole derivatives (), were isolated from the Raphani Semen. Among the indole derivatives, was identified as a new natural product, and , , , , , were isolated from the genus of for the first time. Their structures were elucidated based on the NMR and HR-EI-MS analysis.
