Extraction of roots of Patrinia rupestris (Pall.) Juss. gave a new iridoid compound, 1β,3α-diethyloxy-7-hydromethyl-4-(3-methyl-butyryloxymethyl)-cyclopenta-4(4a),7(7a)-diene[c]pyran-6-one (1), together with a known compound, (1α,4aα, 6α,7β,7aα)-[4a,5,6,7,7a-hexahydro-6,7-dihydroxy-1-(3-methyl-1-oxobutoxy) cyclopenta[c]pyran-4,7-diyl]bis(methylene) 3-methyl-butanoic acid ester (2). The structure of 1 was characterised by HRESIMS, IR, UV, 1-D NMR and 2-D NMR methods. Compound 2 was isolated from this genus for the first time.

Four unreported pyridine alkaloids, curviflorines A-D (1-4), two undescribed iridoids, curviridoids A and B (5 & 6), and one known iridoid glycoside (7), were isolated from the twigs and leaves of Phlogacanthus curviflorus. The structures of these compounds were established by detailed interpretation of MS and NMR data, with the absolute configurations being assigned via comparison of experimental and calculated electronic circular dichroism spectra. Notably, it is the first report of alkaloidal constituents (1-4) from the genus Phlogacanthus.

Enzymes capable of processing a variety of compounds enable plants to adapt to diverse environmental conditions. PRISEs (progesterone-5β-reductase/iridoid synthase-like enzymes), examples of such substrate-promiscuous enzymes, are involved in iridoid and cardenolide pathways and demonstrate notable substrate promiscuity by reducing the activated C=C double bonds of plant-borne and exogenous 1,4-enones. In this study, we identified PRISE genes in () and (), and the corresponding enzymes were determined to share a sequence identity of 95%.

In this work, the first review paper about -iridoids was presented. In particular, their detailed occurrence, chemophenetic evaluation and biological activities were reported. To the best of our knowledge, two hundred and eighty-eight -iridoids have been evidenced so far, bearing different structural features, with the link between two -iridoids sub-units as the major one.
