Minoxidil (2,4-diamino-6-(piperidin-1'-yl)pyrimidine N(3)-oxide; CASRN 38304-91-5) is a bioactive molecule with several nitrosatable groups widely used as an antihypertensive and antialopecia agent. Here the nitrosation of minoxidil was investigated. The conclusions drawn are as follows: (i) In the pH = 2.3-5.0 range, the minoxidil molecule undergoes aromatic C-nitrosation by nitrite. The dominant reaction was C-5 nitrosation through a mechanism that appears to consist of an electrophilic attack on the nitrosatable substrate by H(2)NO(2)(+)/NO(+), followed by a slow proton transfer; (ii) the reactivity of minoxidil as a C-nitrosatable substrate proved to be 7-fold greater than that of phenol, this being attributed to the preferred para- and ortho-orientations of the two -NH(2) groups at positions 2 and 4 of the minoxidil molecule, which activate electrophilic substitution in the C-5 position through their mesomeric effect. The N-nitrosominoxidil resulting from the nitrosation could be potentially harmful to the minoxidil users.

The syntheses of the title compounds demonstrate a privileged introduction of a nitroso (and a hydroxyl via the Baudisch reaction) group to an aromatic ring. These complexes first appeared in the literature as early as 1939, and a range of applications has subsequently been published. However, optimisations of the preparative sequences were not considered, and as such, the reactions have seldom been utilised in recent years; indeed, there remains confusion in the literature as to how such complexes form.

A binuclear Cu(II) complex, 1, [Cu(L)(OAc)](OAc) of imidazole-based ligand LH {LH = 2-(bis(2-ethyl-5-methyl-1H-imidazol-4-yl)methyl)phenol} was synthesized and characterized spectroscopically and structurally. Addition of an equivalent amount of nitric oxide (NO) by a gastight syringe to the acetonitrile:methanol (5:1, v/v) solution of complex 1 at room temperature resulted in the reduction of Cu(II) center to Cu(I) with concomitant C-nitrosation of the ligand. Spectroscopic characterization of the resulting Cu(I) complex (1a) of the C-nitrosylated ligand, L' {L' = 2-(bis(2-ethyl-5-methyl-1H-imidazol-4-yl)methyl)-4-nitroso-phenol} has been done.

Copper(II) complex of 4-amino-3-hydroxy-1-sulphonic acid was synthesized and characterized. Upon addition of nitric oxide, the copper(II) center of the complex in methanol was found to undergo reduction through an unstable copper(II)-nitrosyl intermediate. The formation of the intermediate was confirmed by UV-visible and FT-IR spectroscopy.

Although C-nitroso aromatic compounds have several bioactivities of medicinal interest, the biosynthetic enzymes involved in C-nitrosation have remained unknown until now. Here, we report the entire biosynthesis pathway of 4-hydroxy-3-nitrosobenzamide in Streptomyces murayamaensis, in which a tyrosinase-like copper-containing monooxygenase is responsible for the C-nitrosation. This finding indicates diverse catalytic functions of tyrosinase-like copper-containing monooxygenases in nature.
