Photoaging protective effect of enzyme extracted pomegranate peel against oxidative damage in UVB-irradiated HaCaT cells.

Biomed Pharmacother

Department of Integrated Biomedical and Life Science, Graduate School, Korea University, Seoul 02841, Republic of Korea. Electronic address:

In this study, the ultraviolet B (UVB)-induced skin photoaging inhibitory activity of pomegranate peel extract with increased ellagic acid content through enzymatic hydrolysis was evaluated in HaCaT cells. Among various enzymes, Viscozyme with high tannase and β-glucosidase activities was used, and 1.0 % Viscozyme was added to hydrolyze pomegranate peel for 2 h at 40°C to establish the optimal reaction conditions for high ellagic acid content.

Article Synopsis
  • * MAF demonstrated promising effects in animal and cell models, showing significant reductions in AD symptoms by decreasing inflammation markers and improving skin barrier functions through the modulation of specific cytokines and protein pathways.
  • * The results suggest that MAF may be a viable treatment option for atopic dermatitis, with the ability to alleviate pruritus and enhance skin health, indicating its therapeutic potential.

Article Synopsis
  • - Atopic dermatitis (AD) is a chronic skin condition characterized by inflammation and itchiness, with house dust mites, specifically Der p 38, identified as significant allergens that intensify its development.
  • - In laboratory studies, Der p 38 was found to compromise filaggrin production, a key protein for skin barrier function, while promoting inflammation through various cellular pathways.
  • - Experiments in mice showed that exposure to Der p 38 increased skin lesions and inflammatory cell activity, but these effects were less pronounced in mice lacking the TLR4 receptor, suggesting TLR4's crucial role in the allergenic response triggered by Der p 38.

Structural characterization of human peptidyl-arginine deiminase type III by X-ray crystallography.

Acta Crystallogr F Struct Biol Commun

October 2021

School of Biological Sciences, The University of Auckland, 3a Symonds Street, Auckland 1142, New Zealand.

The Ca-dependent enzyme peptidyl-arginine deiminase type III (PAD3) catalyses the deimination of arginine residues to form citrulline residues in proteins such as keratin, filaggrin and trichohyalin. This is an important post-translation modification that is required for normal hair and skin formation in follicles and keratocytes. The structure of apo human PAD3 was determined by X-ray crystallography to a resolution of 2.

is a beneficial edible plant with a number of biological activities like anti-inflammatory, anti-oxidant, antithrombic, antihyperglycemic, and anti-ischemic activities. The mechanisms behind the antiphotoaging and anti-inflammatory effects of are not fully understood. To investigate how an ethanol extract of (Oe-EE) exerts these effects, we explored its activities in human keratinocytes and dermal fibroblasts.
