Multivalent cyclic RGD conjugates for targeted delivery of small interfering RNA.

We have designed, synthesized, and tested conjugates of chemically modified luciferase siRNA (Luc-siRNA) with bi-, tri-, and tetravalent cyclic(arginine-glycine-aspartic) (cRGD) peptides that selectively bind to the αvβ3 integrin. The cellular uptake, subcellular distribution, and pharmacological effects of the cRGD-conjugated Luc-siRNAs compared to those of unconjugated controls were examined using a luciferase reporter cassette stably transfected into αvβ3 positive M21(+) human melanoma cells. The M21(+) cells exhibited receptor-mediated uptake of cRGD-siRNA conjugates but not of unconjugated control siRNA. The fluorophore-tagged cRGD-siRNA conjugates were taken up by a caveolar endocytotic route and primarily accumulated in cytosolic vesicles. The bi-, tri-, and tetravalent cRGD conjugates were taken up by M21(+) cells to approximately the same degree. However, there were notable differences in their pharmacological effectiveness. The tri- and tetravalent versions produced progressive, dose-dependent reductions in the level of luciferase expression, while the bivalent version had little effect. The basis for this divergence of uptake and effect is currently unclear. Nonetheless, the high selectivity and substantial "knock down" effects of the multivalent cRGD-siRNA conjugates suggest that this targeting and delivery strategy deserves further exploration.

Hydrothermal conversion of actinide oxalates has recently gained attention as an innovative fabrication route for nuclear fuels but has remained mainly limited to tetra- or tri-valent cations. We report herein the reductive conversion of mixtures of uranyl and oxalate ions into UO oxides under mild hydrothermal conditions ( = 250 °C). A multi-parametric study first led to specifying the optimal conditions in terms of pH, oxalate/U ratio and duration to provide a quantitative precipitation of uranium in the hyper-stoichiometric dioxide form with pH = 0.

The effects of simulated radiolytic degradation of tri-butyl phosphate (TBP) on the chemical speciation of cerium were studied by spectrophotometry and electrochemistry of TBP solutions containing increasing amounts of di--butyl phosphoric acid (HDBP), a common degradation product of TBP. Tetravalent cerium was found to exchange coordinated nitrate for the dibutyl phosphate anion, forming dinuclear complexes of the formula (CeOCe)(NO)(DBP)·3TBP ( = 0-3). Compared to Ce(IV), Ce(III) was complexed less strongly by HDBP in TBP, but HDBP displaced both nitrate and TBP to form the series of mononuclear complexes Ce(NO)(HDBP·DBP)·(3-)TBP ( = 0-3).

Tetravalent Cerium Alkyl and Benzyl Complexes.

High-valent cerium complexes of alkyl and benzyl ligands are unprecedented due to the incompatibility of the typically highly oxidizing Ce ion and the reducing alkyl or benzyl ligand. Herein we report the synthesis and isolation of the first tetravalent cerium alkyl and benzyl complexes supported by the tri--butyl imidophosphorane ligand, [NP(Bu)]. The Ce monoiodide complex, [CeI(NP(-butyl))] (), serves as a precursor to the alkyl and benzyl complexes, [Ce(Npt)(NP(-butyl))] () (Npt = neopentyl, CHC(CH)) and [Ce(Bn)(NP(-butyl))] () (Bn = benzyl, CHPh).
