This study presents a selective method of isolation of zearalenone (ZON) and its metabolite, α-zearalenol (α-ZOL), in neoplastically changed human tissue by accelerated solvent and ultrasonic extractions using a mixture of acetonitrile/water (84/16% v/v) as the extraction solvent. Extraction effectiveness was determined through the selection of parameters (composition of the solvent mixture, temperature, pressure, number of cycles) with tissue contamination at the level of nanograms per gram. The produced acetonitrile/water extracts were purified, and analytes were enriched in columns packed with homemade molecularly imprinted polymers. Purified extracts were determined by liquid chromatography (LC) coupled with different detection systems (diode array detection--DAD and mass spectrometry--MS) involving the Ascentis RP-Amide as a stationary phase and gradient elution. The combination of UE-MISPE-LC (ultrasonic extraction--molecularly imprinted solid-phase extraction--liquid chromatography) produced high (R≈95-98%) and repeatable (RSD<3%) recovery values for ZON and α-ZOL.

Mycotoxin exposure from contaminated food is a significant global health issue, particularly among vulnerable children. Given limited data on mycotoxin exposure among Namibian children, this study investigated mycotoxin types and levels in foods, evaluated dietary mycotoxin exposure from processed cereal foods in children under age five from rural households in Oshana region, Namibia. Mycotoxins in cereal-based food samples (n = 162) (mahangu flour (n = 35), sorghum flour (n = 13), mahangu thin/thick porridge (n = 54), oshikundu (n = 56), and omungome (n = 4)) were determined by liquid chromatography-tandem mass spectrometry.

Zearalenone (ZEN) is a harmful macrolide mycotoxin, posing a serious hazard to human health. In this study, a highly efficient ZEN-degrading bacterium Gordonia hydrophobica HAU421 was isolated from soil by using spiramycin (SPM)-containing selective medium. Mass spectrometry analysis revealed that strain HAU421 could transform ZEN into hydrolyzed zearalenone (HZEN), zearalenol (ZEL), and hydrolyzed zearalenol (HZEL).

Most mitigation approaches focus on detecting mycotoxins with standard methods based on liquid chromatography coupled with mass spectrometry (LC-MS). Zearalenone has a high level of detection and exceedance in cereals and by-products. Herein, an electrochemical aptasensor for ZEN detection was proposed. The selected aptamer, which has a high affinity for ZEN, serves as a molecular recognition element and effectively avoids interference from other toxins.

Co-occurrence of multiple mycotoxins is a growing global food safety concern due to their harmful effects on humans and animals. This study developed an eco-friendly sample preparation method and an innovative multiplex microarray-based lateral flow immunoassay, using a novel portable reader for on-site simultaneous determination of five regulated mycotoxins-aflatoxin B, T-2 toxin, zearalenone, deoxynivalenol, and fumonisin B in rice. The eco-friendly and ultrafast extraction procedure utilizes a bio-based solvent.
