The development of disease may be characterized as a pathological shift of homeostasis; the main goal of contemporary drug treatment is, therefore, to return the pathological homeostasis back to the normal physiological range. From the view point of systems biology, homeostasis emerges from the interactions within the network of biomolecules (e.g. DNA, mRNA, proteins), and, hence, understanding how drugs impact upon the entire network should improve their efficacy at returning the network (body) to physiological homeostasis. Large, mechanism-based computer models, such as the anticipated human whole body models (silicon or virtual human), may help in the development of such network-targeting drugs. Using the philosophical concept of weak and strong emergence, we shall here take a more general look at the paradigm of network-targeting drugs, and propose our approaches to scale the strength of strong emergence. We apply these approaches to several biological examples and demonstrate their utility to reveal principles of bio-modeling. We discuss this in the perspective of building the silicon human.

Choline is an important molecule in monitoring food safety and infant nutrition. Here, we report Ce nanogels synthesized by atom transfer radical polymerization (ATRP) employing Ce-coordinated acryloyl-lysine polymer brushes (Ce@SiO NGs) as highly efficient cascade nanozymes for colorimetric detection of choline. The synthesized Ce@SiO NGs demonstrated remarkable peroxidase-like activity with a porous exterior, which are essential to entrap choline oxidase (COx) to yield COx@Ce@SiO NGs and construct a cascade reaction system to detect choline.

Conventional wearable flexible sensing systems typically comprise three components: a flexible substrate that contacts the skin, a signal processing module, and a signal output module. These components function relatively independently, resulting in a complex system that lacks sufficient integration. The work describes a novel sensing and transportation feasibility of the well-established antifungal drug Flucytosine (5-FC) using a 2D Silicon carbide (SiC) and Germanium-doped Silicon carbide (Ge@SiC) nanosheet via PBE level of Density functional theory. The computational study revealed that the drug molecules adhere to SiC and Ge@SiC sheets, maintaining their structural properties through physisorption on SiC and chemisorption on Ge@SiC. The charge transfer process associated with the adsorption is observed by Lowdin charge analysis and both the SiC and Ge@SiC sheets are identified as a feasible oxidation-based nanosensor for the drug.
