The last decade has seen the development of automated microscopy and its adaptation for various areas of research, particularly infectious disease. Most of the high-content screening (HCS) platforms now integrate all of the following necessary steps: automated pipettes for assay miniaturization in 384-well plates, automated image acquisition and data storage and analysis. HCS was initially associated with RNA interference genetic screens for identifying host factors involved in host-pathogen interactions. More recently, both in academia and in industry, HCS has been adapted for drug discovery purposes. High-content analysis enables intracellular tracking of viral particles to profile the antiviral mechanisms of each compound. Adaptation to high-throughput screening in bacteriology and parasitology has already led to the discovery of new types of host-specific inhibitors that differ from those inhibitors that act directly on microbes.

Significance: Imaging flow cytometry allows highly informative multi-point cell analysis for biological assays and medical diagnosis. Rapid processing of the imaged cells during flow allows real-time classification and sorting of the cells. Off-axis holography enables imaging flow cytometry without chemical cell staining but requires digital processing to the optical path delay profile for each frame before the cells can be classified, which slows down the overall processing throughput.

Porcine reproductive and respiratory syndrome virus (PRRSV) causes reproductive failure in sows and respiratory disease in growing pigs, leading to significant economic losses worldwide. Due to the constant mutation and recombination, PRRSV exhibits significant genetic diversity, the general detection of all PRRSV-2 and PRRSV-1 strains is thus needed. In our study, four monoclonal antibodies (mAbs) against PRRSV nucleocapsid (N) protein were generated and the precise and novel B cell epitopes (KPHF and HHTVR) were identified.

Many forms of bioimage analysis involve the detection of objects and their outlines. In the context of microscopy-based high-throughput drug and genomic screening and even in smaller scale microscopy experiments, the objects that most often need to be detected are cells. In order to develop and benchmark algorithms and neural networks that can perform this task, high-quality datasets with annotated cell outlines are needed.

Biodegradable polymeric coatings are being explored as a preventive strategy for orthopaedic device-related infection. In this study, titanium surfaces (Ti) were coated with poly-D,L-lactide (PDLLA, (P)), polyethylene-glycol poly-D,L-lactide PEGylated-PDLLA, (PP20)), or multi-layered PEGylated-PDLLA (M), with or without 1 % silver sulfadiazine. The aim was to evaluate their cytocompatibility, resistance to biofilm formation, and their potential to enhance the susceptibility of any biofilm formed to antibiotics.
