Superoxide Dismutase Activity Measurement Using Cytochrome c-Modified Electrode.

SOD activity was quantified by the use of a cytochrome c-modified gold electrode. The electrode responded rapidly to superoxide radicals in solution. Steady-state superoxide concentrations were established by control of the calibration conditions. On this basis very low SOD activities were detected (10-200 munits/mL). This method showed good correlation with the standard photometric test and was applied for the determination of SOD activity entrapped into liposomes. Interference by hydrogen peroxide and uric acid was characterized and minimized using long-chain thiols for the first electrode modification step. The complete modification proved to be stable for several days.

Protein therapeutics are of widespread interest due to their successful performance in the current pharmaceutical and medical fields, even though their broad applications have been hindered by the lack of an efficient intracellular delivery approach. Herein, we fabricated an active-targeted dual pH-responsive delivery system with favorable tumor cell entry augmented by extracellular pH-triggered charge reversal and tumor receptor targeting and pH-controlled endosomal release in a traceless fashion. As a traceable model protein, the enhanced green fluorescent protein (eGFP) bearing a nuclear localization signal was covalently coupled with a pH-labile traceless azidomethyl-methylmaleic anhydride (AzMMMan) linker followed by functionalization with different molar equivalents of two dibenzocyclooctyne-octa-arginine-cysteine (DBCO-RC)-modified moieties: polyethylene glycol (PEG)-GE11 peptide for epidermal growth factor receptor-mediated targeting and melittin for endosomal escape.

Molsidomine is currently used as a vasodilator drug for the treatment of myocardial ischemic syndrome and congestive heart failure, although still presenting some mitochondrial-targeted side effects in many human cells. As a model of molsidomine mitotoxicity, the reaction of cytochrome c with phosphatidylserine (PS)- and cardiolipin (CL)-containing liposomes was investigated in oxidative/nitrosative conditions imposed by SIN-1 decomposition, which renders peroxynitrite (ONOO) as a main reactive product. In these conditions, the production of thiobarbituric acid-reactive substance (TBARs) and LOOH was affected by the lipid composition and the oxidative/nitrative conditions used.

A cytochrome c (Cyt c)-modified glass nanopore sensing platform was constructed to sensitively detect glucose in single cells based on changes in the ionic current rectification (ICR) of the system. A difference in glucose content between single H8 and HeLa cells in their satiety or starvation states was clearly able to be detected using this method.

A new amperometric biosensor for hydrogen peroxide (HO) and superoxide anion (SO) has been developed. The biosensor developed uses cytochrome c (Cyt c) modified glassy carbon electrodes coupled with electrochemically reduced graphene oxide (ErGO). To immobilize Cyt c, the "one step" electrochemical deposition of vertically aligned and ErGO (VAErGO) has been performed by using a pulse reverse technique, thus resulting in a very simple and efficient system.

In this study a novel cyctochrome c modified nanocomposite electrochemical biosensor was developed for the electrochemical determination of rebaudioside A in different food samples. The electrode surface was fabricated with graphene oxide assimilated with gold nanoparticles decorated on multiwalled carbon nanotubes/cytochrome c. The developed biosensor exhibited a 10-fold enhancement in the differential pulse voltammetry signal carried out at pH 11.
