Addition of neutral R123 molecules (10(-7) M) to an as-prepared gold nanoparticles (AuNPs) suspension generated flocculates that are a small number of closely adjacent particles. Formation of AuNP flocculates was evidenced by the coupled localized plasmon peak at 720-750 nm. The AuNP flocculates provided pronounced SERS spectra of adsorbed neutral R123 molecules (SERS-A) as anticipated by FDTD (Finite Difference Time Domain) simulations. The observed SERS spectra are significantly different from those of cationic R123(+) molecules (SERS-B), which electrostatically adsorbed on Cl(-)-treated AuNPs. The difference is not simply due to deprotonation but reflects a distinct difference in adsorption nature between neutral R123 and cationic R123(+) molecules. Indeed neutral R123 molecules exclusively gave an Au-N stretching band at 202 cm(-1), showing the chemisorption on Au surfaces through lone pair electrons at the amino groups. The different adsorption nature is further evidenced by the observation that cationic R123(+) molecules adsorbed on as-prepared (without NaCl addition) AuNP flocculates gave both SERS-A and SERS-B spectra. Thus, the cationic R123(+) molecules form the flocculates both by chemisorption and electrostatic adsorption owing to modest surface charge on as-prepared AuNPs.

The dynamics and structural characteristics of polymethacrylic acid bound rhodamine-123 (PMAA-R123) and its interpolymer complex formed through hydrogen bonding between the monomeric units with poly(vinylpyrrolidone) were investigated using single molecular fluorescence studies. Dye-exchange dynamics in micellar solutions studied by fluorescence correlation spectroscopy.

We investigated the dye-exchange dynamics between rhodamine 123 (R123), a mitochondrial fluorescent dye, and micelles as membrane mimetic systems. In the presence of neutral micelles (Triton X-100 and Brij 35) R123 partitions between the aqueous solution and the micellar pseudo-phase, undergoing red shift of the absorption and the emission spectra. Fluorescence correlation spectroscopy (FCS) was used to study the dynamics of these systems over an extremely wide time range and at the single-molecule level, yielding information in one and the same experiment about the diffusional dynamics of free and bound rhodamine and about the dye-exchange dynamics as well as several photophysical properties of the rhodamine bound to the micelles.

In this study we demonstrate that haptoglobin, a serum glycoprotein secreted by the liver, has altered structure in the BB/Wor diabetic rat. SDS-PAGE of haptoglobin (a tetramer composed of two glycosylated beta-chains each containing two sites for Asn-linked oligosaccharides connected by disulfide bonds with two nonglycosylated alpha-chains) clearly shows that the beta-chain of haptoglobin from diabetic rats is smaller than normal, with a molecular mass of 39 instead of 40 kDa. Both acute and chronic diabetic rats exhibit the defect.
