Purpose: To improve the mapping of myelin water fraction (MWF) despite the presence of measurement noise, and to increase the visibility of fine structures in MWF maps.
Materials And Methods: An anisotropic diffusion filter (ADF) was effectively combined with a spatially regularized nonnegative least squares algorithm (srNNLS) for robust MWF mapping. Synthetic data simulations were performed to assess the effectiveness of this new method. Experimental measurements of signal decay curves were obtained and MWF maps were estimated using the new method and compared with maps estimated using other methods.
Results: MWF mapping was substantially improved in both simulations and experimental data when ADF was combined with the srNNLS algorithm. MWF variability decreased with the use of the proposed method, which in turn resulted in increased visibility of small focal lesions and structures in the MWF maps.
Conclusion: This study demonstrates that the benefits of ADF and srNNLS algorithms can be effectively combined in a synergic way for robust mapping of MWF in the presence of noise. Substantial improvements to MWF mapping can be made using the proposed method.

Purpose: Myelin quantification is used in the study of demyelination in neurodegenerative diseases. A novel noninvasive MRI method, Microstructure-Informed Myelin Mapping (MIMM), is proposed to quantify the myelin volume fraction (MVF) from a routine multi-gradient echo sequence (mGRE) using a multiscale biophysical signal model of the effects of microstructural myelin and iron.
Theory And Methods: In MIMM, the effects of myelin are modeled based on the Hollow Cylinder Fiber Model accounting for anisotropy, while iron is considered as an isotropic paramagnetic point source.

Purpose: To compare the myelin water fraction (MWF) measurements between 3 T and 7 T and between in vivo and ex vivo human brains, and to investigate the relationship between multi-echo gradient-echo (mGRE)-based 3D MWF and myelin content using histological staining, which has not been validated in the human brain.
Methods: In this study, we performed 3D mGRE-based MWF measurements on five ex vivo human brain hemispheres and five healthy volunteers at 3 T and 7 T with 1 mm isotropic resolution. The data were fitted with the based on a three compartment complex-valued model to estimate MWF.

Non-invasive myelin water fraction (MWF) and g-ratio mapping using microstructural MRI have the potential to offer critical insights into brain microstructure and our understanding of neuroplasticity and neuroinflammation. By leveraging a unique panel of variably hypomyelinating mouse strains, we validated a high-resolution, model-free image reconstruction method for whole-brain MWF mapping. Further, by employing a bipolar gradient echo MRI sequence, we achieved high spatial resolution and robust mapping of MWF and g-ratio across the whole mouse brain.

A new MRI technique is presented for three-dimensional fast simultaneous whole brain mapping of myelin water fraction (MWF), T, proton density (PD), R*, magnetic susceptibility (QSM), and B transmit field (B). Phantom and human (N = 9) datasets were acquired using a dual-flip-angle blipped multi-gradient-echo (DFA-mGRE) sequence with a stack-of-stars (SOS) trajectory.
