Background: DNA damage tolerance (DDT) enables replication to continue in the presence of fork stalling lesions. In mammalian cells, DDT is regulated by two independent pathways, controlled by the polymerase REV1 and ubiquitinated PCNA, respectively.
Results: To determine the molecular and genomic impact of a global DDT defect, we studied Pcna;Rev1 compound mutants in mouse cells.

Background: Anorexia nervosa (AN) is a polygenic, severe metabopsychiatric disorder with poorly understood aetiology. Eight significant loci have been identified by genome-wide association studies (GWAS) and single nucleotide polymorphism (SNP)-based heritability was estimated to be ~ 11-17, yet causal variants remain elusive. It is therefore important to define the full spectrum of genetic variants in the wider regions surrounding these significantly associated loci.

Background/aim: Regulatory T cells (Tregs) play a crucial role in inflammatory responses by regulating the activity of various immune cells. M2 macrophages induced by IL-10 and TGF-β exhibit anti-inflammatory functions and induce Treg differentiation. Although the beneficial effects of 3-bromo-4,5-dihydroxybenzaldehyde (BDB) on various diseases have been widely reported, the mechanisms, through which it alleviates allergic contact dermatitis (ACD) via Tregs and macrophages, are not well understood.

Background/aim: Hederagenin (3β,4α-3,23-dihydroxyolean-12-en-28-oic acid) is a natural pentacyclic triterpene that is present in various medicinal plants and exhibits pharmacological activities against various diseases, including cancer. The aim of the study was to investigate the effect of Aq3639 (3β-[(O-α-L-rhamnopyranosyl-(1→2)-α-L-arabinopyranosyl)oxy]olean-12-en-28-oic acid), a hederagenin glycoside comprising hederagenin and a disaccharide of L-rhamnose and L-arabinose, on breast cancer cells.
Materials And Methods: Aq3639 was isolated from the pericarps of Akebia quinata fruits, and its effects on cells from the human breast cell line MCF-7 were examined.
