Published: November 1994

122 human uterine leiomyomas, representing the main chromosomal aberrations seen in these tumours, were analyzed by Southern blotting. Probes for the four oncogenes, gli, fos, jun, and met, all localized in or close to the chromosomal breakpoints in leiomyomas, were used. In only one single leiomyoma, with a 7p+q- marker, was there evidence of rearrangement of one of the oncogenes, namely met.

PDE4 inhibitor rolipram represses hedgehog signaling via ubiquitin-mediated proteolysis of GLI transcription factors to regress breast cancer.

Aberrant activation of the hedgehog (Hh) signaling pathway positively correlates with progression, invasion and metastasis of several cancers, including breast cancer. Although numerous inhibitors of the Hh signaling pathway are available, several oncogenic mutations of key components of the pathway, including Smoothened (Smo), have limited their capability to be developed as putative anti-cancer drugs. In this study, we have modulated the Hh signaling pathway in breast cancer using a specific FDA-approved phosphodiesterase 4 (PDE4) inhibitor rolipram.

Stereocaulon alpinum has been found to have potential pharmaceutical properties due to the presence of secondary metabolites such as usnic acid, atranorin, and lobaric acid (LA) which have anticancer activity. On the other hand, the effect of LA on the stemness potential of colorectal cancer (CRC) cells remains unexplored, and has not yet been thoroughly investigated. In this study, we examined the inhibitory activity of LA from Stereocaulon alpinum against the stemness potential of CRC cells and investigated the possible underlying mechanisms.

The functional activation of the androgen receptor (AR) and its interplay with the aberrant Hh/Gli cascade are pivotal in the progression of castration-resistant prostate cancer (CRPC) and resistance to AR-targeted therapies. Our study unveiled a novel role of the truncated form of Gli (t-Gli3) in advancing CRPC. Investigation into Gli3 regulation revealed a Smo-independent mechanism for its activation.

[Mortality due to mesothelioma and asbestosis in Campania Region (Southern Italy): perspectives for reducing asbestos exposure].

Epidemiol Prev

December 2024

Dipartimento di Medicina, Epidemiologia, Igiene del lavoro e ambientale, Istituto Nazionale per l'Assicurazione contro gli Infortuni sul Lavoro, Roma.

Objectives: to provide an overview of the geographical distribution of mesothelioma and asbestosis deaths in the Campania Region (Southern Italy) occurred from 2005 to 2018 and to identify areas at higher risk.

Design: for each municipality, Standardized Mortality Ratios (SMRs) for mesothelioma and asbestosis have been estimated from the mortality data provided by the Italian National Institute of Statistics (Istat). Deaths for which mesothelioma and asbestosis were identified as the underlying causes, according to the classification system ICD-10 codes (C45 and J61, respectively), were included.

Article Synopsis
  • Breast cancer, especially triple-negative breast cancer (TNBC), presents a significant challenge due to its aggressive nature and poor prognosis, prompting research into potential treatments like pirfenidone, an FDA-approved drug with anti-fibrotic and emerging anti-tumor properties.
  • In experiments, pirfenidone was shown to reduce cell viability and proliferation in TNBC cell lines (MDA-MB-231 and HCC-1937), shifting more cells into the G0/G1 phase of the cell cycle and inducing apoptosis by altering specific protein expressions related to cell survival and apoptosis.
  • The study revealed that pirfenidone's effectiveness is linked to its suppression of the Hedgehog/GLI1 signaling pathway, as
