Crystal structure and biophysical characterisation of Helicobacter pylori phosphopantetheine adenylyltransferase.

Helicobacter pylori is a bacterium that causes chronic active gastritis and peptic ulcers. Drugs targeting H. pylori phosphopantetheine adenylyltransferase (HpPPAT), which is involved in CoA biosynthesis, may be useful. Herein, we report the expression in Escherichia coli and purification of recombinant HpPPAT and describe a crystal structure for an HpPPAT/CoA complex. As is the case for E. coli PPAT (EcPPAT), HpPPAT is hexameric in solution and as a crystal. Each protomer has a well-packed dinucleotide-binding fold in which CoA binds. Structural characterisation demonstrated that CoA derived from the E. coli expression system bound tightly to HpPPAT, presumably to initiate feedback inhibition. However, the interactions between the active-site residues of HpPPAT and CoA are not identical to those of other PPATs. Finally, CoA binding affects HpPPAT thermal denaturation.

Developing high-performance and low-cost electrodes for hydrogen and oxygen evolution reactions (HER and OER, respectively) represents a pivotal challenge in the field of water electrolysis. Herein, W doped NiFe LDH nanosheets (NiFe-W/NF) were immobilized on nickel foam (NF) through one-step corrosion engineering, which induced the coexistence of α-Ni(OH) and β-Ni(OH). The doping of large atomic radius W influenced the growth of crystal planes of Ni(OH), promoting the formation of α-Ni(OH), which results in large layer spaces and neatly arranged nanosheets structure.

Bagaza virus (BAGV) is a mosquito-borne flavivirus and has caused significant avian death in many regions, and also garnered recognition as a significant human pathogen causing diseases like encephalitis. Numerous compounds with useful biological activities were previously prepared by tethering ferrocene to natural product scaffolds. Some conjugates of ferrocene and steroids in particular were demonstrated to act as both potent and selective antiproliferative agents. Motivated by a lack of structural diversity in the pregnane series of conjugates, we performed chemical modifications of several naturally occurring progestogens, their biosynthetic precursors and chemically related derivatives so as to obtain conjugates 1-8, in which the steroid skeleton and ferrocene core are linked by the steroid sidechain.
