Semaphorin3A (Sema3A) is a repulsive guidance molecule for axons, which acts by inducing growth cone collapse through phosphorylation of CRMP2 (collapsin response mediator protein 2). Here, we show a role for CRMP2 oxidation and thioredoxin (TRX) in the regulation of CRMP2 phosphorylation and growth cone collapse. Sema3A stimulation generated hydrogen peroxide (H2O2) through MICAL (molecule interacting with CasL) and oxidized CRMP2, enabling it to form a disulfide-linked homodimer through cysteine-504. Oxidized CRMP2 then formed a transient disulfide-linked complex with TRX, which stimulated CRMP2 phosphorylation by glycogen synthase kinase-3, leading to growth cone collapse. We also reconstituted oxidation-dependent phosphorylation of CRMP2 in vitro, using a limited set of purified proteins. Our results not only clarify the importance of H2O2 and CRMP2 oxidation in Sema3A-induced growth cone collapse but also indicate an unappreciated role for TRX in linking CRMP2 oxidation to phosphorylation.

The current work presents the flame-retardant performance of hybrid polypropylene composites, reinforced with specific short woven flax fabrics (SWFs), short basalt fibers (BFs), and rice husk powder (RHP), using polypropylene grafted maleic anhydride (MAPP) as the coupling agent. Horizontal burning test (HBT), microcalorimeter test (MCT), and cone calorimeter test (CCT) were conducted on these composites. The formulations used were 25% SWF/PP, 25% SWF/20% BF/PP, and 25% SWF/20% BF/PP with 6% RHP and 25% SWF/20% BF/PP with varying RHP contents (6, 12, and 18%) in combination with 6% MAPP.

Signal transduction downstream of axon guidance molecules is essential to steer developing axons. Second messengers including cAMP are key molecules shared by a multitude of signaling pathways and are required for a wide range of cellular processes including axon pathfinding. Yet, how these signaling molecules achieve specificity for each of their downstream pathways remains elusive.

The aim of this study was to explore the factors influencing the quality of new bone formation after distraction osteogenesis in children with Pierre Robin sequence (PRS). Using cone-beam computed tomography (CBCT), bone density relative grayscale values of the region of new bone formation before and 3 to 4 months after mandibular distraction osteogenesis (MDO) were measured in 80 children with PRS, and correlation analysis was conducted with the potential clinical influencing factors of the children. CBCT reconstruction of the panoramic film showed that the new bone formation was good at 3 to 4 months after MDO.

The issue of hazardous smoke and toxic gases released from epoxy resins (EP), which often causes casualties in real fires, has limited its application. Therefore, we have developed a novel flame retardant based on a bimetallic-doped phytate-melamine (BPM) structure with Zn and Fe ions incorporated into the polymer matrix using a straightforward solution-based synthetic method. The combustion performance of the composite was evaluated using a cone calorimeter test, which showed that the peak heat release, total heat release, and total smoke production were reduced by 50%, 31.

Connectome generative models, otherwise known as generative network models, provide insight into the wiring principles underpinning brain network organization. While these models can approximate numerous statistical properties of empirical networks, they typically fail to explicitly characterize an important contributor to brain organization-axonal growth. Emulating the chemoaffinity-guided axonal growth, we provide a novel generative model in which axons dynamically steer the direction of propagation based on distance-dependent chemoattractive forces acting on their growth cones.
