We develop a fluorescent ruthenium metalloglycocluster for use as a powerful molecular probe in evaluating the binding between carbohydrates and lectins by fluorescence emission (FE) and fluorescence polarization (FP) analyses. Changes in the FE and FP of these metalloglycoclusters are measured following the addition of lectin [peanut agglutinin (PNA), Ricinus communis agglutinin 120, Concanavalin A (ConA), or wheat germ agglutinin] or tetanus toxin c-fragment (TCF). After the addition of PNA, the FE spectrum of [Ru(bpy-2Gal)(3)] shows a new emission peak and the FP value of [Ru(bpy-2Gal)(3)] increases. Similarly, the FE spectrum of [Ru(bpy-2Glc)(3)] shows a new emission peak and the FP value increases on addition of ConA. Because other combinations of metalloglycoclusters and lectins show little change, specific binding of galactose to PNA and that of glucose to ConA are confirmed by the FE and FP measurements. Resulting dissociation constants (K(d)) prove that the metalloglycoclusters with highly clustered carbohydrates show higher affinity for the respective lectins than those with less clustered carbohydrates. Furthermore, specific binding of [Ru(bpy-2Gal)(3)] to TCF was confirmed by the FP measurement.

Novel coumarin-triphenyliminophosphorane (TPIPP) fluorophores, synthesized via a nonhydrolytic Staudinger reaction, exhibit remarkable redox-responsive optical properties. Upon chemical and electrochemical oxidation, these compounds display a hypsochromic shift in absorption from 430 to 350 nm, accompanied by up to 11-fold fluorescence enhancement under 405 nm excitation. In 2001, Tang's team discovered a unique type of luminogens with substantial enhanced fluorescence upon aggregation and introduced the concept of "aggregation-induced emission (AIE)". Unlike conventional fluorescent materials, AIE luminogens (AIEgens) emit weak or no fluorescence in solution but become highly fluorescent in aggregated or solid states, due to a mechanism known as restriction of intramolecular motions (RIM). Initially considered a purely inorganic chemical phenomenon, AIE was later applied in biomedicine to improve the sensitivity of immunoassays.

The extensive use of tetracyclines in livestock poses health risks due to their residues in animal-derived food; therefore, developing simple detection methods to replace complex traditional approaches is of paramount importance. Here, we developed a dual-ligand zinc-based metal-organic framework material, Zn-BTC-BDC-NH (denoted as ZTD), for the detection of tetracyclines. The intrinsic blue fluorescence of ZTD was quenched upon the introduction of tetracyclines due to electron transfer from -NH of ZTD to -CO- and -OH groups of tetracycline molecules; meanwhile, the new green fluorescence emission was generated through π-π stacking between aromatic rings and the formation of complexes between Zn and C-O/C═O groups.
