Two new glycoconjugate vaccines protecting against invasive diseases caused by 13 pneumococcal serotypes or 4 meningococcal serogroups will soon be available in Switzerland. They offer real new perspectives for young children, and for high-risk group adults. As their development is recent, the data one may wish to have is not yet fully available for each age and disease categories. This article attempts to delineate the interest of these new glycoconjugate vaccines and their limits. The additional information (and the answers to numerous expected questions) will be distributed to the 4300 subscribers of InfoVac (www.infovac. ch), which just celebrated its 10th birthday.

Group A Streptococcus (Strep A) is a human-exclusive bacterial pathogen killing annually more than 500,000 patients, and no current licensed vaccine exists. Strep A bacteria are highly diverse, but all produce an essential, abundant, and conserved surface carbohydrate, the Group A Carbohydrate, which contains a rhamnose polysaccharide (RhaPS) backbone. RhaPS is a validated universal vaccine candidate in a glycoconjugate prepared by chemical conjugation of the native carbohydrate to a carrier protein.

Human leukocyte antigen (HLA)-G is a nonclassical HLA class I molecule that has an immunosuppressive effect mediated by binding to immune inhibitory leukocyte immunoglobulin-like receptors (LILR) B1 and LILRB2. A conventional HLA-G isoform, HLA-G1, forms a heterotrimeric complex composed of a heavy chain (α1-α3 domains), β2-microglobulin (β2m) and a cognate peptide. One of the other isoforms, HLA-G2, lacks a α2 domain or β2m to form a nondisulfide-linked homodimer, and its ectodomain specifically binds to LILRB2 expressed in human monocytes, macrophages, and dendritic cells.

Bacterial surface polysaccharides are widely decorated with diverse rare functional groups. An understanding of the role of these functional groups in immunological activity of the glycans is required for the development of carbohydrate-based treatment against bacterial infectious diseases. Here, the antigenicity evaluation of synthetic trisaccharide 1 related to Plesiomonas shigelloides serotype O51 lipopolysaccharide (LPS) O-antigen and its acetamido derivative 2 was achieved to elucidate the immunological importance of O-antigen structure.
