The genome sequence for Schistosoma mansoni has been determined, allowing the complete protein complement to be predicted. However, few functional genomics techniques have been developed for use in S. mansoni, limiting the usefulness of the sequence data. Here we describe a whole mount in situ hybridization (WISH) method that can be used to identify the tissue-specific expression of transcripts in S. mansoni. Using this protocol we determine the tissue-specific expression of tetraspanin 2, a female-enriched tetraspanin, phenol oxidase, the secretory Cu/Zn superoxide dismutase, and an Argonaute family member. The localization of these transcripts by WISH correlates with prior studies performed using immunohistochemistry and/or in situ hybridization on tissue sections. WISH can be adapted to screen multiple transcripts, thus identifying novel targets for drugs or vaccines.

In this article, we report the first case of a 61-year-old woman who was diagnosed with both nodules and cystic lesions in her lungs. The lung nodules were diagnosed as ALK-positive histiocytosis (APH) carrying an gene fusion, which microscopically displayed a mixed morphology of foamy cells, spindle cells, and Touton's giant cells. Immunohistochemistry showed expression of CD163, CD68, and ALK, while fluorescence hybridization (FISH) with second-generation sequencing (NGS) showed the ALK gene fusion with the FLCN gene variant.

The 2018 ASCO/CAP guidelines for HER2 testing for breast cancer implemented the addition of immunohistochemistry (IHC) directed in situ hybridization (ISH) recount to resolve equivocal results. The implementation of an additional 2+ IHC-directed ISH recount adds additional complexity to the testing workflow for an unclear impact on HER2 results. A retrospective review of all equivocal ISH cases (groups 2, 3, and 4) that underwent 2+ IHC-directed ISH, since the 2018 guidelines, which were finalized as either amplified or not amplified, was performed.
