[Molecular recognition elements--DNA/RNA-aptamers to proteins]. | LitMetric

In this review summarizes data on DNA/RNA aptamers--a novel class of molecular recognition elements. Special attention is paid to the aptamers to proteins involved into pathogenesis of wide spread human diseases. These include aptamers to serine protease, to cytokines/growth factors, to influenza viral protein, nucleic acid binding proteins. Strong and specific binding for a given protein target of aptamers make them an attractive class of direct protein inhibitors. They can inhibit pathogenic proteins and it is becoming clear that aptamers have the potential to be a new and effective class of therapeutic molecules.

Duck viral hepatitis severely threatens the development of the duck industry, leading to economic losses every year. Using selected Pekin duck populations exhibiting varying resistance towards Duck Hepatitis A Virus type 3 (DHAV-3), screening for genetic variations, such as single nucleotide polymorphisms (SNP), associated with disease susceptibility will facilitate the breeding of Pekin ducks with enhanced disease resistance. The biological role of fucosyltransferases, which are a type of glycosyltransferase enzymes, is to catalyze the transfer of fucose to molecules such as oligosaccharides, glycoproteins and glycolipids, which is crucial for maintaining immune function by promoting effective pathogen recognition and modulating immune responses through specific fucosylation patterns.

Chagas disease, caused by the protozoan Trypanosoma cruzi, remains a major public health issue, particularly in Latin America, and is traditionally transmitted by triatomine bugs. Chagas disease progresses through two phases: an acute phase characterized by high parasitemia and a chronic phase, which may remain asymptomatic or develop into severe complications. This study utilized dual-RNAseq technology to analyze gene expression in mice fibroblast host cells infected with the strain JJ21 isolated from an acute Chagas disease case of presumptive oral transmission, comparing it with the MG strain isolated from a chronic Chagas disease human case.

Selective recognition of Zn in biological and environmental samples is a challenge. An N, N-diethyl substituted coumarin thiosemicarbazone was designed as a fluorogenic sensor, HADICIT, [(E)-2-(1-(7-(diethylamino)-2-oxo-2H-chromen-3-yl)ethylidene)-N-isopropylhy drazine-1-carbothioamide] for the detection of Zn to address the issue. The compound was characterized using FTIR, H NMR, C NMR, and mass spectrometry methods.

Organophosphate pesticides (OPs) are widely used in agricultural fields and can inhibit the activity of human acetylcholinesterase (hAChE) by covalently binding to serine at the enzyme's active site. However, the molecular recognition mechanisms beyond their covalent binding remain unclear. This study employed molecular docking along with molecular dynamics simulations (MD) to investigate four representative OPs, Phosphamidon, Monocrotophos, Dichlorvos, and Trichlorfon, as well as two potential alternatives Magnolol (MAG) and Honokiol (HON), to understand the conformational change of hAChE and its molecular recognition mechanism.

Structural basis of thymidine-rich DNA recognition by Drosophila P75 PWWP domain.

Drosophila P75 (dP75), a homolog of the human LEDGF/p75, is crucial for oogenesis by recruiting the histone kinase Jil-1 to euchromatin and impeding H3K9me2 spreading. Like LEDGF, dP75 binds transcriptionally active chromatin, but its precise mechanism remains unclear. Here we show that its PWWP domain prefers binding to thymidine-rich DNA over GC-rich sequences.
